摘要
随着荧光定量PCR技术广泛应用,内参基因稳定性作为影响定量实验结果准确性的关键因素而逐渐成为学者研究热点。植物候选内参基因稳定性评价已有诸多研究报道,候选内参基因的选择也不再局限于传统的管家基因,基因芯片及转录组测序技术发展也使一些新的基因作为候选内参基因成为可能,有学者也选择mi RNA作为候选内参基因。而评价这些候选内参基因的方法也由原来单一的软件评价,发展为结合各个不同的软件综合分析,使内参基因的评价更加可靠、准确。本综述主要研究了近几年来药用植物候选内参基因的选择、稳定性分析等研究进展,旨在为后续开展药用植物育种相关研究提供一定的参考与借鉴。
With the development of real-time quantitative PCR(RTq-PCR), the selection of reference genes is crucial for the results of relative quantification analysis of gene expression. There have many reports about the stability analysis of candidate reference genes. Not only the traditional housekeeping genes, but also some new genes and mi RNA based on the transcription analysis were used for candidate reference genes. Several evaluation methodologies were used for stability analysis comprehensively, that enhanced the reliability and veracity of the selection of reference genes. The application of reference genes in medicinal plant in recent years were reviewed,in order to provide the reference for the relative studies on the medicinal plant genetic engineering in the future.
