农杆菌介导茶树遗传转化中抗生素种类和浓度优化 被引量:7

Studies on Optimization of Antibiotic Species and Concentration in Genetic Transformation of Camellia sinensis L. Mediated by Agrobacterium

摘要 茶树是一种多年生木本植物,建立其基因转化体系是进行基因工程育种的重要环节。在以茶树体胚为材料,研究了三种抗生素对其抑菌和生长的影响。结果表明,三种抗生素对茶树体胚的抑菌和生长有明显的不同,随着抗生素浓度的不同,茶树体胚生长趋势明显不同;头孢霉素400 mg/L与潮霉素15 mg/L协同抑制农杆菌,促进茶树体胚的增殖;与其相比,特美汀300 mg/L抑制农杆菌,显著促进茶树体胚的增殖,有植物激素的作用。本研究为下一步茶树的遗传转化奠定基础。 Tea plant is a perennial woody plants, the establishment of its genetic transformation system is an important part in the genetic engineering breeding. In this study, the tea plant embryos as materials, and the effects of three kinds of antibiotics on the antibacterial and growth were studied. The results showed that three kinds of antibiotics had different effects on the antibacterial and growth of tea plant embryos. With different concentration of antibiotic, tea plant embryo growth trend was obviously different. It is showed that 400 mg/L toxin cephalosporin and 15 mg/L hygromycin synergistic inhibition of agrobacterium and promoted the tea plant embryo proliferation; In contrast, 300 mg/L Timentin inhibition of agrobacterium, significantly promoted the tea plant embryo proliferation and had effect on plant hormones. This research had a basis foundation for further study of genetic transformation in tea plant.
