The Study on Regeneration In Vitro of White Flower Chinese Kale

摘要 以白花芥蓝品种"明丰香菇"为试验材料,系统研究了不同消毒组合处理对种子发芽的影响,不同激素配比与外植体类型对不定芽分化及再生的影响。分析可得:酒精15 s+5%NaClO 6 min消毒处理效果最好;带柄子叶的愈伤发生率和不定芽分化频率均高于下胚轴,其中带柄子叶的最优激素组合为0.03 mg/L NAA+0.75 mg/L 6-BA,愈伤发生率为98.3%,不定芽分化率达80%,再生系数为3.03;分化的不定芽在添加0.03 mg/L NAA的MS培养基上生根效果最好,生根率达100%,形成的不定根多且粗壮。试管苗移栽成活率高达93%。试验结果为芥蓝遗传转化研究提供了依据和参考。 In the present study, Chinese kale cultivar"mingfengxianggu"was chosen as plant material, the effects of sterilizing treatment on seed germination, different hormones combinations and explant types on shoot formation and plant regeneration were investigated. The results showed that the optimal sterilizing treatment was75% ethanol and 5% NaCl O together, and the sterilizing time were 15 seconds and 6 minutes, respectively. The frequencies of callus formation and shoot differentiation from cotyledon with petiole were both higher than those of hypocotyl. The best combination was cultured on MS medium supplied with 0.03 mg/L NAA and 0.75 mg/L6-BA, while the highest rates of callus formation and shoot differentiation from cotyledon with petiole were up to98.3% and 80%, respectively, and the regeneration factor reached 3.03. The MS medium with 0.03 mg/L NAA was found to be suitable for rooting culture, which with 100% of rooting rate, more and stronger roots were induced.The transplanting survival rate was 93%. The results provided the basis and reference for genetic transformation of Chinese kale.
