当归对大鼠胶原性关节炎滑膜MCP-1、MMP-3影响及病理改变 被引量:4

The Effect of Danggui on MCP-1 and MMP-3 in Collagen Induced Arthritis Synovium and the Pathologic Change

摘要 目的:观察当归对胶原性关节炎(CIA)模型大鼠关节滑膜单核细胞趋化因子-1(MCP-1)、基质金属蛋白酶-3(MMP-3)的影响及病理改变。方法:将清洁级Wistar大鼠30只随机分为空白对照组,胶原性关节炎(CIA)空白组,CIA甲氨蝶呤组,CIA当归低剂量组和CIA当归高剂量组,每组6只。采用Ⅱ型胶原免疫法制备胶原性关节炎模型,制模成功后分别给药。甲氨蝶呤组每次给药1mg/kg,每周2次,腹腔注射;当归组采用灌饲,分别为低剂量组0.75g/d和高剂量组0.75g×2/d,每天应用。所有药物均使用3周。观察3周后大鼠滑膜MCP-1、MMP-3的变化规律及大鼠关节滑膜的病理改变特点。结果:与空白对照组比较,CIA空白组,CIA甲氨蝶呤组,CIA当归低剂量组和CIA当归高剂量组的大鼠滑膜MCP-1、MMP-3显著增高。与CIA空白组比较,CIA甲氨蝶呤组,CIA当归低剂量组和CIA当归高剂量组的大鼠滑膜MCP-1均有降低;CIA甲氨蝶呤组,CIA当归低剂量组和CIA当归高剂量组的大鼠滑膜MMP-3均有降低。CIA空白组典型的滑膜组织病理改变为细胞核呈蓝色,胞浆为淡红色,视野内可见典型的炎性肉芽肿,内可见炎症细胞浸润,巨噬细胞和多核巨细胞等,组织水肿明显;当归高剂量组部分有典型的炎性肉芽肿,但炎性细胞浸润相对偏少,组织水肿较对照组减轻,而低剂量组的病理改变基本同对照组,变化不明显。结论:当归能不同程度降低胶原性关节炎大鼠滑膜MCP-1、MMP-3的含量,呈现剂量依赖性改变;当归能够减轻胶原性关节炎大鼠滑膜的典型病理改变,减少肉芽肿生成,减轻组织水肿。 Objective: To observe the effect of Angelica sinensis on the expression of MMP-3 and MMP-3 in rats with collagen-induced arthritis (CIA). Methods: 30 Wistar rats were randomly divided into control group, collagen group (CIA) group, CIA methotrexate group, CIA angelica low dose group and CIA angelica high dose group, 6 rats in each group. The collagen type arthritis model was prepared by collagen type 11 immunization method. Methotrexate group was given lmg/kg twice a week for intraperitoneal injection. Angelica group was fed with 0.75g/d low dose group and 0.75g × 2/d in high dose group, respectively. All drugs were used for 3 weeks. The changes of MCP-1 and MMP-3 in rat synovium and the pathological changes of rat synovium were observed after 3 weeks. Results: Compared with the control group, the levels of MCP-I and MMP-3 in the synovium of CIA group, CIA methotrexate group, CIA angelica low dose group and CIA angelica high dose group were significantly higher. Compared with CIA blank group, MCP-1 in CIA methotrexate group, CIA angelica low dose group and CIA angelica high dose group were decreased. CIA metho- trexate group, CIA angelica low dose group and CIA The high levels of MMP-3 in rat high-dose group were decreased. CIA blank group typical synovial histopathological changes in the nucleus was blue, the cytoplasm was light red, visible visual inflammatory granuloma, visible inflammatory cell infiltration, macrophages and multinucleated giant cells, tis-sue edema significantly There was a typical inflammatory granuloma in the high dose group, but the infiltration of in- flammatory cells was relatively less, tissue edema was less than that of the control group, and the pathological changes of the low dose group were basically the same as the control group. Conclusion: Angelica sinensis can decrease the con- tent of MCP-1 and MMP-3 in synovium of collagen-induced arthritis rats in a dose-dependent manner. Angelica sin- ensis can reduce the typical pathological changes of synovium in rats with collagenous arthritis and reduce granuloma Generate, reduce tissue edema.
