生长因子EGF、aFGF、bFGF对毛乳头细胞生长的研究 被引量:3

摘要 目的研究生长因子EGF、a FGF、b FGF对体外培养人毛乳头细胞生长的调控作用。方法获取人头皮外伤缝合术多余头皮组织,一步酶消化法获取人毛乳头细胞并将其贴壁培养,分为实验组和对照组。实验组在培养基中添加生长因子,而对照组培养基则不添加。采用细胞生物学技术,免疫荧光技术,CCK8细胞增殖实验,研究生长因子EGF、a FGF、b FGF对体外培养的毛乳头细胞增殖与干性表达影响。结果在人毛乳头细胞体外培养中,实验组细胞生长比对照组增殖更加明显(P<0.05),干细胞阳性率更高。结论生长因子EGF、a FGF、b FGF能促进毛乳头细胞生长和维持干细胞特性。 Objective To study the regulating effect of the growth factor EGF, aFGF, bFGF to the human dermal papilla ceils. Methods To get the redundant scalp tissue from the human scalp trauma suture, one step enzyme digestion to obtain the dermal papilla and adherent culture, divided into the experimental group and the control group. The experimental group culture medium added the growth factor, however, the control group did not exist. Using the cell biology technology, immunofluorescence technique and CCK8 cell proliferation experiment, to study the impact of the growth factor EGF, aFGF, bFGF to the dermal papilla cells" proliferation and stem expression. Results In the culture of the dermal papilla cells in vitro, the experimental group proliferation more obvious than the control group (P〈 0. 05), stem cell positive rate higher. Conclusions The growth factor including EGF, aFGF, bFGF can promote the dermal papilla cells growth and maintenance of stem cell characteristics.
