蛋氨酸对水貂毛乳头细胞生长的影响 被引量:4

摘要 为了研究在培养基中添加不同浓度蛋氨酸对水貂毛乳头细胞增殖、周期的影响。使用两步酶法分离培养毛乳头细胞,经鉴定后,在对照组常规培养液中分别添加不同浓度的蛋氨酸,通过MTT法计算毛乳头细胞的增殖率、流式细胞术检测蛋氨酸对细胞周期的影响。结果培养基中添加蛋氨酸可以促进细胞增殖,其中添加8μg/m L蛋氨酸组的增殖率最高;当浓度64μg/m L时,对毛乳头细胞的增殖有抑制作用。添加8μg/m L蛋氨酸组处于G1期的细胞比率小于对照组,细胞增殖指数、S期和G2期比率明显高于其他两组。表明在本试验条件下,在培养基中添加适宜浓度的蛋氨酸可以促进水貂毛乳头细胞的增殖,提高细胞的增殖活力。 We studied the effect of different methionine level on proliferation and cycle of mink dermal papilla cells (DPCs). DPCs were isolated and cultivated with two step method. After cells were identified, the fourth passage cells were cuhured in medium supplemented with methionine. The cell proliferation was analyzed by MTT, and flow cytometry was used to evaluate the effect of Met on DPC cycles. Our results showed that methionine enhanced DPC proliferation, especially the addition of 8μg/mL of methionine. However, it suppressed the proliferation of DPC at the concentration of 64μg/mL. Cells in GI phase in the group with 8μg/mL methionine were less than that in the control group. However,and eeU proliferation index,and cells in S and G2 phases were significantly higher than the other two groups. Adding appropriate concentration of methionine in the culture medium can promote DPC proliferation and enhance cell proliferation activity.
