恒河猴源犬瘟热病毒N、F基因的克隆与序列分析 被引量:2

摘要 为了进一步明确引起恒河猴类麻疹综合征的病原体及其来源,对恒河猴源犬瘟热病毒的N、F全基因进行了克隆和序列分析。得到全长1 572 bp的N基因和2 080 bp的F基因,测序后分别与不同CDV毒株的N、F基因核苷酸和氨基酸序列进行分析,N基因核苷酸(氨基酸)与CDV疫苗株Onderstepoort、vaccine x、标准野毒株A75-17同源性分别为93.6%(96.9%)、97.3%(99.0%)和97.3%(99.0%);F基因核苷酸(氨基酸)与上述毒株同源性分别为91.0%(97.8%)、94.9%(98.6%)和94.4%(97.9%)。N、F基因的系统发育分析表明,恒河猴CDV毒株与黑龙江狐狸毒株(CDV-FOX-HLJNM)遗传关系较近,与其他分离株遗传关系相对较远。本研究从分子水平上证明了引起恒河猴发病的病原体为一株CDV野毒株,而非疫苗株。 The full-length N and F genes of CDV in rhesus monkey were cloned and sequenced. The full length of N gene and F gene was 1572 hp and 2080 hp,respeetively. The results indicated that the homology of the nucleotides (and amino acids) of the N gene with CDV vaccine Ondersteponrt vaeeine x and standard wild strain A75-17 were 93.6% (96. 9% ) , 97.3% (99.0%) and 97.3% ( 99. 0% ) , respectively. The homology of the nueleotides( and amino acids) of the F gene with CDV vaccine Ondersteport and vaccine x aml staudard wild strain A75-17 were 91.0% (97. 8% ) ,94. 9% (98.6%)and 94.4% (97.9%) ,respectively. The phylogenie analysis showed that the CDV of rhesus monkey had close relationship with Fox strain of Heilungiiang( CDV - FOX - HIJNM ). The resuh proved thai the CDV in rhesus monkey was a wiht strain,but not a vaccine strain.
