摘要
目的探讨类风湿关节炎(RA)患者外周血单个核细胞体外诱导成树突状细胞(DC)TIM4表达。方法分离15例RA患者和10例正常对照外周血单核细胞,加入重组人巨噬集落形成因子(rhMCSF)、重组人白介素-4(rh IL-4)培养6 d,再加入肿瘤坏死因子-α(TNF-α)培养1 d。采用流式细胞仪检测DC诱导分化情况,实时荧光定量PCR和Western blot检测TIM4表达。结果 RA患者外周血单核细胞诱导分化成DC数量多于对照组(P<0.01),DC表达TIM4明显增加(P<0.01)。结论 DC表达TIM4上调可能参与RA发病。
Objective To study TIM4 expression of in vitro induced dendritic cells (DCs) form peripheral blood mononuclear cells (PBMCs) in patients with rheumatoid arthritis (RA). Methods PBMCs were isolated from 15 RA patients and 10 healthy controls, and then cultured with recombinant human macrophage colony stimulating factor (rhMCSF) and rhlL-4 for 6 d, and with additional tumor necrosis factor ct (TNF-α) for 1 d. The induced DCs were determined by flow cytometry, and TIM4 expression was detected by real-time fluorescent quantitative PCR and Western blot. Results The percentage of DCs and expression of TIM4 were higher in RA group than in control group (P〈0.01). Conclusion TIM4 overexpression of DCs might be implicated in the pathogenesis of RA.
