摘要
目的探讨雪莲培养细胞提取物对辐射所致小鼠造血功能损伤的影响。方法100只昆明种雄性小鼠,采用数字随机表法平均分为空白对照组、单纯辐射组、雪莲培养细胞提取物高剂量组(1.0g/kg)、中剂量组(O.5g/kg)、低剂量组(0.1g/kg),每组20只。雪莲培养细胞提取物给予相应剂量的雪莲培养细胞提取物灌胃,其他各组给予蒸馏水灌胃,连续28d。除空白对照组外,其余各组于灌胃14d后给予2.5Gy 60Coγ射线全身辐射。辐射前第14天、l天及辐射后第3、6、10、14天检测外周血白细胞数,辐射后第6、14天检测脾脏指数、内源性脾结节数、骨髓有核细胞数和骨髓DNA含量。结果各组小鼠体重在饲喂过程中均明显增加(t=76.58~203.34,P〈0.01);与空白对照组比较,单纯辐射组小鼠外周血白细胞数、骨髓DNA含量、脾脏指数均明显下降(P〈0.01),脾结节数均明显增加(P〈O.01);与单纯辐射组相比,雪莲培养细胞提取物各组小鼠外周血白细胞数、骨髓DNA含量和骨髓有核细胞数均明显升高(P〈O.05),脾脏指数与内源性脾结节数增加(P〈0.05),检测结果不呈剂量依赖性。结论雪莲培养细胞提取物对辐射致造血系统损伤小鼠有修复和促再生作用。
Objective To explore the effects of Saussurea involucrata cultured cell extracts on hematopoietic function in the mice with radiation injuries. Methods By using digital random table method, 100 Kunming male mice were averagely divided into, 5 groups: blank control group, pure radiation group, Saussurea involucrata cultured cell extracts high, medium and low dosage group. The high, medium and low dosage group were respectively administrated Saussurea involucrata cultured cellextracts with the dosages of 1.0 g/kg, 0.5 g/kg, 0.1 g/kg and the others were given distilled water for 28 days. Except blank control group, the other groups were given 2.5 Gy 60Co 7 ray irradiation after daily intragastric administration for 14 days. The number of peripheral blood leukocytes in the mice was respectively checked 14 d and 1 d before irradiation and 3 d, 6 d, 10 d and 14 d after irradiation. The spleen index, the endogenous splenic nodule count, the bone marrow nucleated cells count and the bone marrow DNA content were respectively checked 10 d and 14 d after irradiation. Results Mice weight was all raised during administration in 4 dosage groups (t=76.58-203.34, P〈0. 01). Compared with those of blank group, the number of peripheral blood leukocytes, the bone marrow DNA content and the spleen index of pure radiation group were significantly decreased (P〈0.01), but the endogenous splenic nodules significantly increased (P〈0.01). Compared with those of pure radiation group, the number of peripheral blood leukocytes, the bone marrow DNA content, the spleen index and the endogenous splenic nodule count of Saussurea involucrata cultured cell extracts groups were significantly increased (P〈0.05). The spleen index and endogenous splenic nodules were significantly increased (P〈0.05). The result was not in a dose- dependent manner. Conclusions Saussurea involucrata cultured cell extracts had the effect of injury repair and regeneration on hematopoietic system of radiation injured mice.
