HPLC法测定玉葡萄根中儿茶素的含量

Determination Catechin of Root of Ampelopsis Delavayana by HPLC

目的:建立高效液相色谱法定量测定玉葡萄根中儿茶素。方法:玉葡萄根甲醇提取液,采用Agilent ZORBAX Extend-C_(18)柱(4.6mm×250mm,5μm),流动相为乙腈-0.1%磷酸水溶液(10:90);体积流量:1.0mL/min;柱温:30℃;检测波长:280nm。结果:儿茶素在0.1998~1.9980μg(r=0.9997)范围内具有良好的线性关系,平均回收率为99.32%,RSD为0.83%。结论:本方法操作简便,结果准确,可以较好的测定儿茶素。

Objective To establish a method for determining catechin of root of Ampelopsis delavayana by HPLC.Methods Theanalysis of A.delavayana methanolic extract was carried out on Agilent ZORBAX Extend-C18 column（4.6mmx250mm,5μm）with acetonitrile-0.1% phosphoric acid aqueous （10：90）as mobile phase,flow rate wsa 1.0mL/min,column temperature was maintained at 30℃,and detection wavelength was set at 280 nm.Results catechin was 0.1998-1.9980μg（r=0.9997）,respectively,The average recoveries were 99.32%（RSD 0.83%）.Conclusion The method is simple,accurate and candeterminecatechin.
