EGFR突变肺癌中长链非编码RNA表达谱的生物信息学分析及高表达lncRNA CR749465的验证 被引量:3

摘要 目的:探讨及分析EGFR突变肺癌中差异表达长链非编码RNA的表达谱及分析其功能。方法:从NCBI公共数据平台GEO下载肺癌基因芯片数据GSE31210,获得127例EGFR突变肺癌和20例正常组织的基因表达谱数据,R软件包处理获得差异表达的lncRNA。并用荧光定量PCR法检测19对EGFR突变肺癌患者和癌旁标本,验证候选基因lncRNA CR749465的差异表达。利用DAVID分析候选lncRNA相关靶基因的GO分类及pathway通路。结果:GSE31210芯片数据分析显示,127个基因在EGFR突变肺癌组织出现差异表达,其中上调的有96个,下调的有31个。经荧光定量PCR验证,候选基因lncRNA CR749465基因表达情况与芯片数据分析结果一致。结论:EGFR突变肺癌中存在明显的lncRNAs差异性表达,通过GO及KEGG分析发现,CR749465可能在EGFR突变肺癌的发生和发展中发挥重要作用。 ObjectiveTo analyze IncRNA expression profiles in EGFR mutation lung carcinoma and preliminary validation. Methods : The publicly available gene chip data GSE31210, which consists of 127 EGFR mutation lung carcinoma and 20 normal tissues,were downloaded from the GEO of NCBI. 19 pairs of EGFR mutation lung cancer pa-tients, specimens were used to validate the differentially expressed gene by real - time PCR. The interesting candidate IncRNA related mRNA were analyzed with on - line bioinformatics tools DAVID. Results : There were 127 differential-ly -expressed IncRNA in GSE31210, among which 96 were up - regulated and 31 were down - regulated. The expres-sions of IncRNA CR749465 were confirmed by realtime PCR. Conclusion :The results reveal IncRNAs expression pat-terns in EGFR mutation lung carcinoma by microarray. The IncRNA CR749465 may play a key role in tumor develop-ment and occurrence.
