摘要
目的研究槟榔炮制过程中槟榔碱转化途径及美拉德反应对其的影响。方法应用固态反应模型模拟槟榔炮制过程,采用离子交换HPLC法和ESI-MS法测定槟榔碱在美拉德反应时的转化差异,推导美拉德反应对槟榔碱转化的影响及槟榔碱转化的路径。结果槟榔碱在炮制过程中具有两种转化路径:一是槟榔碱的去甲基化和甲基化,二是槟榔碱的缩合反应。在美拉德反应存在时,前者的转化率明显下降。结论槟榔炮制过程中,美拉德反应对槟榔碱去甲基化和甲基化的转化过程具有抑制作用,可使槟榔碱的含量维持在适当水平,保证槟榔炮制品的药理活性。
OBJECTIVE To investigate the arecoline transformation and the impact of Maillard reaction on it during Arecae semen parching process. METHODS Solid state reaction models was applied to simulate Arecae semen parching process. Arecoline transfor- mation paths and the influence of Maillard reaction on it were derived by analyzing the difference of arecoline transformation between the presence and absence of Maillard reaction by ion exchange HPLC and ESI - MS. RESULTS Two paths of arecoline transformation at parching temperatures were established. Path 1 was demethylation and methylation,path 2 was condensation of arecoline. The arecoline transformation in path 1 was declined obviously in the presence of Maillard reaction. CONCLUSION The demethylation and methyla- tion of arecoline are inhibited by Maillard reaction during Arecae semen parching process, so the content of arecoline can reach an appro- priate level to maintain the pharmacological activity.
