摘要
目的考察天门冬酰胺酶(L-Asp)羟丙基-β-环糊精脂质体(AHL)的稳定性及相关机制。方法采用逆相蒸发法制备AHL,对其低温放置稳定性、热稳定性、酸碱稳定性、血浆稳定性、抗胰蛋白酶水解能力及相关机制进行初步研究。结果 AHL体外稳定性都优于L-Asp,能阻止L-Asp构象的改变。结论 AHL能提高L-Asp的稳定性。
OBJECTIVE To investigate the stability and related mechanism of L - asparaginase (L - Asp ) loaded in hydroxypropyl- β- cyclodextrin liposomes (AHL). METHODS The reverse evaporation method was used to prepare AHL. The temperature stability, acid - base stability, plasma stability, proteolytic stability, and related mechanism of AHL were investigated. RESULTS AHL could improve the in vitro stability of L - Asp. AHL might inhibit the conformational changes of L - Asp. CONCLU- SION AHL can improve the stabilities of L - Asp.
