摘要
目的 探讨在不同前列腺组织中斑点型锌指结构蛋白(SPOP)的表达,及其表达与前列腺癌分期、分级的关系.方法 收集我院正常前列腺组织(NP)12例、前列腺增生组织(BPH)12例、前列腺癌旁组织(PC)12例以及前列腺癌组织(PCa)46例.经常规处理后,应用免疫组织化学法检测SPOP表达.结果 PCa组中SPOP表达28例(28/46), NP、PC组表达均为阳性;BPH组11例表达阳性(11/12);PCa组与NP组(P=0.009)、BPH组(P=0.043)及PC组(P=0.009) 比较,差异均有统计学意义.Gleason评分≥8分组(2/11)与2-4分组(9/12, P=0.006)、5-7分组(17/23, P=0.002)比较,SPOP表达阳性率差异有统计学意义. T3(4/11)与T1(6/7)期(P=0.040)、T3(4/11)与T2(14/16)期(P=0.006)、T4(4/12)与T1(6/7)期(P=0.027)、 T4(4/12)与T2(14/16)期(P=0.003)、T3+T4(8/23)与 T1+T2(20/23)期(P=0.000)组间比较差异均有统计学意义.SPOP表达阳性率在转移组(3/12)与非转移组(25/34)比较差异有统计学意义(P=0.003).结论 PCa组织中SPOP表达明显低于其他类型前列腺组织,并与PCa评分、分期明显相关.
Objective To explore the expression of speckle-type poxvirus and zinc finger (POZ) protein (SPOP) in normal prostate (NP), benign prostate hyperplasia (BPH), pericancerous (PC) tissue and prostate cancer (PCa), and study its correlation with the grade and stage of PCa.Methods Totally 12 NP slides, 12 BPH slides, 12 PC slides and 46 PCa slides from our hospital were collected.These slides were routinely processed and analyzed according to the requirement of immunohistochemical staining.Tumors were classified according to the 2002 TNM staging system.The grading system used was based on the Gleason grade.Results We found that the expression of SPOP in PCa (28/46) was significantly lower than that in NP (12/12, P=0.009), BPH (11/12, P=0.009) and PC (12/12, P=0.009) tissue, and the difference was statistically significant (P〈0.05).With regard to grade of PCa, the expression of SPOP in PCa with Gleason score ≥8 group (2/11) was lower than the Gleason score 2-4 group (9/12, P=0.006) and the Gleason score 5-7 group (17/23, P=0.002).The difference was statistically significant (P〈0.05).Moreover, the T3 (4/11) and T4 (4/12) stages had a less positive rate than the T1 (6/7) and T2 (14/16) stages.There was statistically significant difference between early and advanced stage of PCa (P=0.000).Furthermore, the positive expression of SPOP in PCa samples decreased significantly in the metastasis group (3/12, P=0.003).Conclusion SPOP may be related to the development of PCa and the expression of SPOP in PCa correlates with the grade and stage of PCa.
