大孔树脂分离纯化迷迭香叶总黄酮及抗氧化活性研究 被引量:10

摘要 目的:探讨大孔树脂分离纯化迷迭香叶总黄酮及抗氧化活性。方法:选择6种类型大孔树脂,比较其吸附量、吸附率和解吸率,筛选最佳树脂,单因素分析最佳纯化工艺条件,检测迷迭香叶总黄酮体外抗氧化活性。结果:AB-8为最佳树脂,上样液浓度为2.25mg/mL,上样流速为3BV/h,pH为3.15,上样体积为1.5BV。以4BV 80%乙醇在流速2BV/h下洗脱,得黄酮的纯度为68.39%,精制倍数为3.37。迷迭香总黄酮对DPPH和ABTS自由基具有良好的清除能力。结论:AB-8树脂对迷迭香叶总黄酮具有良好的吸附和解吸效果,且迷迭香叶总黄酮具有良好的抗氧化作用。 Objective. Explore the separation, purification and antioxidant activity of total flavonoids from Rosmarinus officinalis L. with maeroporous resin. Methods: Six types of macroporous resins are selected to compare their performances in absorbing and desorbing flavonoids from Rosmarinus officinalis L.. The optimal purification conditions are analyzed by single-factor experiment. Results. The macroporous resin AB-8 is the best, the sample concentration is 2.25 mg/mL, the adsorption rate is 3 BV/h, the pH is 3.15, the sample volume is 1.5 BV, the eluting reagent is 4 BV 80M ethanol, the desorption rate is 2 BV/h, the dose of reagent is 4 times as much as the column volume. The purity of flavonoids is 68. 39%, and the purification factor is 3. 37. The total flavonoids from Rosmarinus officinalis L. have good antioxidant ability to DPPH and ABTS. Conclusion: The results show that AB-8 resin could be used for separation and purification of total flavonoids from Rosmarinus officinalis L. and has good antioxidant ability.
