摘要
目的建立测定柴胡配方颗粒中柴胡皂苷a含量的高效液相色谱(HPLC)法。方法色谱柱采用Welch Ultimate XB-C18柱(250 mm×4.6 mm,5μm),以乙腈-甲醇-0.1%磷酸溶液为流动相进行梯度洗脱,检测波长为210 nm。结果柴胡皂苷a进样量在0.660 4~6.604 0μg范围内与峰面积线性关系良好,回归方程为Y=288.216 X-6.057,r=0.999 97,柴胡皂苷a的平均回收率为101.99%,RSD为1.91%(n=9)。结论该方法简便、准确,重复性好,可用于柴胡配方颗粒的质量控制。
Objective To establish an HPLC method for the content determination of saikosaponin a in Chaihu Dispensing Granules.Methods HPLC analysis was performed on an Welch Ultimate XB-C18 column(250 mm×4.6 mm,5μm) with acetonitrile —methanol — 0.1%phosphoric acid solution as mobile phase for gradient elution.The detection wavelength was at 210 nm.Results The calibration curve of saikosaponin a showed a good linearity within the range of 0.660 4-6.604 0μg,and the regression equation was Y = 288.216X-6.057(r = 0.999 97).The average recovery was 101.99%and RSD was 1.91%(n = 9).Conclusion The method is convenient,accurate and reproducible to operate and is suitable for the quality control of Chaihu Dispensing Granules.
