榴莲皮中总黄酮的提取及其抗氧化活性 被引量:5

摘要 以榴莲皮为原料,添加纤维素酶、表面活性剂提取榴莲皮中的总黄酮.研究了纤维素酶及表面活性剂用量、乙醇浓度、提取温度、提取时间对总黄酮产率的影响,采用Box-Behnken试验设计建立了提取的试验模型及最佳提取条件,并对提取物的抗氧化活性进行了初步试验.在酶用量为20U/mL、表面活性剂用量为5mg/mL、乙醇浓度为30%、提取温度为70℃、提取时间为70min时,总黄酮产率为3.14%.榴莲皮总黄酮具有清除DPPH自由基、超氧阴离子、羟自由基活性能力,清除能力与其浓度具有一定量效关系. With durian peel as the raw materials, flavonoids in durian peel were extracted by adding cellulose and surfac- rant. Influence of cellulase and surfactant dosage, ethanol concentration, extracting temperature, and extracting time on yield of flavonoids, was studied. The extracting experimental models were established and the optimal conditions were found by Box- Behnken experimental design, and the antioxidative property of the extract was investigated preliminarily. When the dosage of enzyme was 20U/mL, and the amount of surfactant was 5mg/mL, ethanol concentration was 30%, extracting temperature was 70℃, and extracting time was 70 min, then the yield of flavonoids would be 3.14%, which has a the relative error of 2. 6% compared with the value of 3.06% through theoretical calculation. The flavonoids in durian peel had the active ability to clear DPPH radicals, superoxide anion, and hydroxyl radicals, and the clearing ability had some dose-effect relationship with the concentration.
