摘要
目的:观察生晒参水煎液对斑马鱼血管生长的作用,为人参的临床应用提供理论依据。方法:选用转基因斑马鱼(fli1a-EGFP),建立正常血管生长模型和内皮细胞生长因子受体酪氨酸激酶抑制剂(VRI)诱导的血管损伤模型,使用不同浓度生晒参水煎液进行干预,分别观察斑马鱼肠下静脉和节间血管的情况。结果:斑马鱼正常血管生长模型中,生晒参水煎液各浓度组与对照组的肠下静脉平均出芽数、平均交叉数和血管直径无明显差异(P>0.05)。斑马鱼血管损伤模型中,与正常组比较,VRI组及人参水煎液各浓度组的完整型血管明显减少,缺陷型血管增多(P<0.01);与VRI组比较,人参水煎液3μg/m L组的完整型血管明显增多(P<0.05),缺陷型血管明显减少(P<0.01),100μg/m L组完整型血管明显减少(P<0.05),余无统计学差异(P>0.05)。结论:人参水煎液不能促进斑马鱼正常血管生长,但在较低浓度时可以抑制VRI诱导的血管损伤,而高浓度时则相反。
Objective:To explore the effect of ginseng water decoction on angiogenesis in zebrafish. Methods:Transgenic zebrafish(fli1a-EGFP)was used to establish angiogenesis model and angiogenesis injury model with VRI. The zebrafish subintestinal vessels(SIVs)and intersegmental vessels(ISVs)were observed after different concentration treatment of ginseng water decoction. Results:In angiogenesis zebrafish model,no significance existsed in average sprouting number,average cross number,or vessels diameter among all groups. In angiogenesis injury zebrafish model,compared with control group,intact vessels were significantly reduced but defective vessels increased in VRI group and all ginseng groups(P〈0.01). Compared with VRI group,intact vessels significantly increased(P〈0.05)and defective vessels decreased(P〈0.01)in 3 μg/m L ginseng group,but in 100 μg/m L ginseng group intact vessels were reduced(P〈0.05). Conclusion:Ginseng water decoction has no effect on simple angiogenesis in zebrafish,but could inhibit angiogenesis injury induced by VRI in low concentration,which is on the contrary in high concentration.
