摘要
目的:观察熏洗I号对大鼠肛瘘术后模型创面表皮细胞生长因子(EGF)表达的影响。方法:SD大鼠40只制成肛瘘术后创面模型,并按随机数表达法分为生理盐水组(10只),创面用生理盐水熏洗;高锰酸钾组(10只),创面用高锰酸钾熏洗;实验组(10只),创面用自拟中药"熏洗I号"熏洗;正常组(10只)。于造模成功后第10天采用Western blot蛋白印迹及免疫组化法观察EGF在创面的表达,作为创面肉芽组织增生情况的指标;造模成功后第1天、第10天从大鼠眼底静脉丛采血检测血液学变化,作为炎症变化指标;测定大鼠创面愈合率,作为判断创面愈合情况的指标;采用光密度法测定4组大鼠EGF的表达量。结果:以愈合率达到95%为创面愈合,实验组创面平均愈合时间15 d,与其它3组对比具有统计学差异。血液学结果提示在术后10 d中药熏洗I号组大鼠的炎症表现得到明显控制。Western blot蛋白印迹及免疫组化法显示,实验组大鼠创面肉芽组中EGF的阳性表达明显高于其它3组(P<0.01)。结论:自制中药洗剂"熏洗I号"可以促进肛周污染性创面释放表皮细胞生长因子(EGF),加速上皮化进程,从而达到促进创面愈合的作用。
Objective:To evaluate the impact of"Fumigation I"on the expression of epidermal growth factor(EGF)in rats with fistulectomy wounds. Methods:A total of 40 SD rats were subjected to fistulectomy,then randomized into four groups each with 10 rats. For each group,the wounds were subjected to either no treatment,or fumigation with one of the following:saline,potassium permanganate,and herbal lotion"Fumigation I". To evaluate proliferation rate of granuloma,EGF expression at the infistulectomy wounds was measured by western blot and immunohistochemistry ten days after surgery. Inflammation was determined by hematological analysis with blood samples from the fundus venous plexus at one day and ten days post-surgery. Wound healing rate was determined by healing at the fistulectomy site in rats. EGF expression of the four groups of rats was determined by light spectrometry. Results:Using 95% recovery rate as the criteria for would healing,the average recovery time for the group treated with herbal lotion"Fumigation I"was 15 days,which was statistically significantly shorter than the other three groups. Hematology analysis showed that inflammation was well controlled in the"Fumigation I"treated group. Western blot and IHC showed EGF expression in granuloma at the infistulectomy wounds was significantly higher than the other three groups as well(P〈0.01). Conclusion:Herbal lotion"Fumigation I"promotes the release of EGF from perianal wounds,accelerates epidermal wound healing,thus facilitates healing of fistulectomy wounds.
