摘要
目的:建立不同批次、不同产地单味药组方的杏苏止咳颗粒高效液相色谱指纹图谱,为建立快速、高效的杏苏止咳颗粒指纹图谱质量控制方法提供实验依据。方法:采用HPLC-DAD法,Thermo Syncronis C_(18)(250 mm×4.6 mm,5μm)色谱柱;乙腈-0.01%甲酸水为流动相;流速为1.0 m L·min^(-1);检测波长为278 nm;柱温为27℃;进样量为20μL,建立杏苏止咳颗粒指纹图谱;通过SPSS 19.0统计软件进行相似度评价,结合聚类分析与主成分分析(PCA),对不同批次的杏苏止咳颗粒进行质量评价。结果:建立了杏苏止咳颗粒指纹图谱共有模式,确定了13个共有峰,18批杏苏止咳颗粒的相似度为0.951~0.998;通过聚类分析可大致聚成3类;PCA结果与聚类分析结果一致。结论:杏苏止咳颗粒的HPLC-DAD指纹图谱的构建和化学模式的识别为杏苏止咳颗粒质量控制提供依据。
Objective :To establish the HPLC fingerprint for effective quality control of Xingsu Zhike Granules. Methods:The fingerprint of Xingsu Zhike Granules was developed with ultra - performance liquid chromatography, and the Thermo Syncronis C18 (250 mm×4.6 mm,5 μm)was used in the gradient elution with a mobile phase of acetonitrile- water (both containing 0.01% fomlic acid). The flow rate was 1.0 mL · min- 1 ;the column temperature was 27℃ and the detection wavelength was at 278 nm. Similarity evaluation combined with hierarchical clustering analysis (HCA) and principal components analysis (PCA) were used to evaluate the quality of Xingsu Zhike Granules by the statistical software of SPSS19.0. Results :The mutual mode of HPLC finger- print of Xingsu Zhike Granules was established with good precision,reproducibility and 13 common peaks in the fingerprint were determined. The similarity of 18 samples was 0.951 -0. 998. The PCA result was consistent with the HCA. Conclusion:The estab- lishment of HPLC - DAD fingerprint of Xingsu Zhike Granules and the application of chemical pattern recognition can provide the reference for the quality control of Xingsu Zhike Granules.
