摘要
目的:探讨葛根素对人子宫颈癌细胞株HeLa的作用及其机制。方法:取对数生长期的HeLa细胞分别用不同剂量的葛根素处理(0μmol,12.5μmol,25μmol,50μmol)。细胞培养48 h后,利用MTT法检测HeLa细胞增殖,流式细胞术检测细胞凋亡水平,RT-PCR法检测Bax mRNA和Bcl-2 mRNA水平。Western blotting法检测Wnt、β-catenin、Bax、Bcl-2、p53和p21表达水平。结果:葛根素成剂量依赖性地抑制HeLa细胞增殖,促进细胞凋亡,促进BaxmRNA表达,减少Wnt、β-catenin、Bcl-2 mRNA、p53和p21表达。结论:葛根素可抑制宫颈癌HeLa细胞增殖并诱导其凋亡,其作用机制与减少Wnt/β-catenin信号通路活化相关。
Objective:To investigate the effect and mechanism of puerarin on human cervical cancer cell line HeLa.Methods:HeLa cells in logarithmic growth phase were treated with different doses of Puerarin(0,12.5,25,50) mol.The proliferation of HeLa cells was detected by MTT assay after cell culture for 48 h,and Flow cytometry was used to detect the level of cell apoptosis.The level of Bax mRNA and Bcl-2 mRNA were detected by RT-PCR and Western blotting was used to detect the expression of Wnt,β-catenin,Bax,Bcl-2,p53 and p21.Results:Puerarin inhibited the proliferation of HeLa cells,promoted the expression of Bax mRNA and decreased the expression of Wnt,β-catenin,Bcl-2 mRNA,p53 and p21,all in dose-dependent manner.Conclusion:Puerarin can induce the proliferation and apoptosis of cervical cancer HeLa cells,the mechanism of which may be related to the anti-activation of Wnt/β-catenin signal pathway.
