摘要
目的:采用细胞培养的方法,体外观察疏肝健脾活血方药物血清对转化生长因子β1(transforming growth factor,TGFβ1)诱导的大鼠肝星状细胞(hepatic stellate cell,HSC-T6)增殖作用的影响。方法:将40只大鼠随机分成正常大鼠血清组,疏肝健脾活血方低剂量组,疏肝健脾活血方中剂量组,疏肝健脾活血方高剂量组。以疏肝健脾活血方给大鼠灌胃,腹主动脉采血并制备血清。采用TGFβ1诱导HSC-T6作为活化的肝星状细胞的研究模型,以不同浓度的疏肝健脾活血方药物血清作用于体外培养的HSC-T6,分别作用24 h、48 h后,采用CCK-8法检测不同浓度的疏肝健脾活血方药物血清对HSC-T6增殖的影响。结果:疏肝健脾活血方药物血清作用于TGFβ1诱导的HSC-T6,细胞增殖受到明显抑制。各组作用48 h后,空白对照组OD值为(0.617±0.061),疏肝健脾活血方低剂量组、中剂量组、高剂量组OD值分别为(0.446±0.091)、0.417±0.103)、(0.376±0.065),疏肝健脾活血方各剂量组与空白组比较,差异均有统计学意义(P<0.05)。正常大鼠含药血清组OD值为(0.506±0.015),与空白对照组比较,差异无统计学意义(P>0.05)。结论:疏肝健脾活血方药物血清对TGFβ1诱导的HSC-T6细胞活化作用增殖有抑制作用。
Objective:To explore the influence of proliferative effects of Shugan Jianpi Huoxue Decoction drug serum on hepatic stellate cells(HSC-T6) induced by transforming growth factor 1(TGFβ1).Methods:40 rats were randomly divided into normal rat serum group;Shugan Jianpi Huoxue Decoction in low,middle and high concentration group.Rats were given intragastric administration of Shugan Jianpi Huoxue Decoction,abdominal aortic blood serum and preparation.Taking TGFβ1 induced activated HSCT6 cells as research model,with different concentrations of Shugan Jianpi Huoxue Decoction drug serum,the influence of proliferative effects of Shugan Jianpi Huoxue Decoction drug serum on HSC-T6 were studied by CCK-8 assay at 24 and 48 hours,respectively.Results:The Shugan Jianpi Huoxue Decoction drug serum had inhibitory effect,in time and concentration dependent,on TGFβ1 induced HSC-T6 cells.Compared with blank control group,the values of A of different groups whose cells were treated by different concentrations of drug sera at low(0.446 ± 0.091),middle(0.417 ± 0.103) and high(0.376 ± 0.065),especially 48 h later,there were significant differences in each concentration group(P 0.05).showed significantly difference(P 0.05).The values of A of normal rat serum group A was(0.506 ± 0.015),compared with the control group,the difference was not statistically significant(P 0.05).Conclusion:The Shugan Jianpi Huoxue Decoction drug serum has certain inhibitory effect on proliferative effect of TGFβ1induced activated HSC-T6 cells.
