摘要 目的:优选茯苓水溶性粗多糖的水提醇沉工艺。方法:以茯苓水溶性多糖提取率为参察指标,以水为溶剂,选取粉碎力度、料液比、提取时间、提取次数、醇沉浓度、浓缩倍数、醇沉时间为影响因素,进行单因素考察,在综合单因素实验的基础上,采用L9(34)正交试验对水提和醇沉工艺进行优化设计,采用硫酸-蒽酮法测定多糖的含量。结果:茯苓水溶性多糖最佳的水提醇沉工艺为:过60目筛、料液比为1∶10,提取时间为100 min,提取1次,提取液浓缩至原体积的1/12,醇沉浓度为80%,静置12 h,过滤,滤饼用体积分数80%乙醇洗涤,60°C减压干燥。结论:此工艺成本低、操作简便、重现性好,可用于茯苓药材及饮片粗多糖的提取。 Objective:To optimize the water extract and ethanol precipitation process of water insoluble polysaccharides from Poria cocos.Methods:On the basis of single factor test with crushing,strength,materia liquid ratio,extraction time,extraction times and alcohol concentration,concentration ratio,alcohol sink for influence factors,the orthogonal test with 4 factors and 3 levels method was used to design and the content of the extract was determined using anthrone-sulfuric acid colorimetry method.Results:The results showed that the optimal extraction for obtaining the highest amounts of polysaccharides was carried out by over 60 mesh,water(1∶10),with refluxing for 100 mins,the filtrat was concentrated to 1/12 V,then,ethanol was added to concentration of 80%,stewing 12 h,the precipitate filtated and washed with 80% ethanol,the product was dried under vacuum at 60° C.Conclusion:The process has low cost,simple operation and good reproducibility.It can be used for the extraction of crude polysaccharides from Poria cocos herbs and Yinpian.
