摘要
研究蛋白的超声辅助提取工艺和体外抗氧化活性。在单因素试验的基础之上根据Box-Behnken中心组合设计原理,借助Design-Expert V 8.0.6软件,建立四因素三水平的响应面模型,研究蛋白提取的料液比、提取时间、超声时间和pH这四个因素及其交互作用对提取率的影响,并以VC为对照,对其还原能力和对DPPH的清除能力进行探讨。结果表明,最佳的蛋白提取工艺参数为:料液比1︰30 g/m L,提取时间36 h,超声时间27 min,pH 10。白骨壤蛋白的提取率达39.23%。得到的蛋白对DPPH的IC_(50)为3.13μg/m L;在试验范围内对DPPH的最大清除率为82.59%,表现出较好的体外抗氧化活性。
Response surface methodology was used to optimize the processing of protein from Avicennia marina. The preparation process of Avicennia marina protein was optimized according to Box-Benhnken central composition design principle on foundation of single factor experiment, the method of response surface analysis with 4 factors and 3 levels combined with Design-Expert V 8.0.6. Data analysis software was adopted to study material/liquid ratio, extraction time, ultrasonic time, and pH four factors and their interactions on extraction effect. According to the above results, the optimum preparation conditions were obtained as followings: Material/liquid ratio 1 : 30 g/mL, extraction time 36 h, ultrasonic time 27 rain, pH 10. The extraction of Avicennia marina protein reached 39.23%. In DPPH radical scavenging assays, the IC50 of Avicennia marina protein was 3.13μg/mL, and the maximum scavenging rates was 82.59%. These results supported strong antioxidant activity of Avicennia marina protein.
