葡萄柚中色素组分的测定及萃取条件对色素的影响 被引量:2

Determination of Pigment Components in Grapefruit and the Effect of Extraction Conditions on the Pigment

摘要 采用高效液相色谱法比较不同提取工艺对葡萄柚(Citrus paradise Macf.)中叶黄素(Lut)、玉米黄素(Zea)、β-隐黄素(β-Cryp)、α-胡萝卜素(α-C)和β-胡萝卜素(β-C)5种色素含量的影响。采用Waters YMCTM Carotenoid3μm色谱柱(4.6 mm×150 mm),流动相A相为乙腈,流动相B相为乙酸乙酯,梯度洗脱,流速1.0 m L/min,DAD检测波长450 nm,柱温30℃。检测方法显示,4种多酚的检测线性范围为0.1~2 000 mg/L;检测限在0.002~0.01 mg/L之间,加标回收率在97.11%~101.02%之间,检测方法灵敏有效。检测结果表明,不同提取工艺得到的葡萄柚中叶黄素、玉米黄素、β-隐黄素、α-胡萝卜素和β-胡萝卜素5种葡萄柚色素的总含量以亚临界萃取最高,且亚临界萃取过程没有加热处理,最大程度地保留了葡萄柚色素。从葡萄柚色素的含量及物质特性保留的角度考虑,选择葡萄柚的提取工艺以亚临界萃取为宜。 The influence of different extraction conditions on contents of 5 kinds of the pigments including lutein (Lut), zeaxanthin (Zea),β-cryptoxanthin (β-Cryp), a-carotene (a-C) and fl-carotene (β-C) in grapefruit was compared by using HPLC. The experiment was based on Waters YMCTM Carotenoid 3μm column (4.6 mm×150 mm) with mobile phase A of acetonitrile and mobile phase B of ethyl acetate, and gradient elution with the flow rate of 1.0 mL/min, DAD detection wavelength of 450 nm and column temperature of 30 %. Detection method showed sensitive and effective; Four kinds of polyphenols linear range were 0.1-2 000 mg/L; The detection limit was between 0.002-0.01 mg/L; The recoveries were between 97.11%-101.02%. The results that the total content of five pigments in grapefruit was the highest when using subcritical extraction among all the extraction methods, due to no heating treatment during subcritical extraction, which could keep the pigments in grapefruit by maximum. It was the best to choose subcritical extraction to extract the grapefruit, from the aspects of increasing pigment content and retaining material properties.
