摘要 目的建立高效液相色谱-蒸发光散射检测器(HPLC-ELSD)法测定六神曲中的青蒿素的含量。方法采用Kromasil C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.2%乙酸(62∶38)为流动相进行洗脱,流速1.0 m L·min^(-1),柱温25℃;蒸发光散射检测器漂移管温度50℃,载气压力30 psi,增益值为50;进样量10μL。结果青蒿素在0.5~10.0μg线性关系良好,R2=0.9992。方法回收率为96.7%,RSD=3.8%(n=6)。结论本法具有良好的精密度和重现性,结果准确可靠,可作为六神曲饮片的质量控制方法,为生产提供依据。 Objective To develop an HPLC-ELSD method for the determination of artemisinin in Liushenqu. Methods The separation was performed on a Kromasil C18 column (250 mm×4.6 mm, 5 μm), and acetoni- trile-0.2% acetic acid (62 : 38) was used as the mobile phase. The flow was 1.0 mL · min ^-1, and the tempera- ture was 25 ℃. The temperature of detection was 50 ~C. The pressure of N2 was 30 psi, the gain value was 50, and the injection volume was 10 μL. Results The calibration curve showed a good linearity at 0.5 - 10.0 μg (R2 = 0.9992), the recovery was 96.7%, and RSD was 3.8% (n = 6). Conclusion The method is accurate and reproducible, and can be used for the determination of artemisinin in Liushenqu.
