戒烟联合N-乙酰半胱氨酸改善烟熏大鼠肺损伤研究 被引量:3

摘要 目的观察戒烟和N-乙酰半胱氨酸(NAC)干预对大鼠肺组织形态的影响以及血浆同型半胱氨酸(HCY)水平的变化。方法将雄性Wistar大鼠50只分为对照组、烟熏组、烟熏+NAC组、烟熏+戒烟组、烟熏+NAC+戒烟组,每组各10只。5组大鼠均在实验前及8周时进行大鼠内眦静脉采血,离心后ELISA法检测大鼠血浆HCY水平。HE染色观察大鼠肺组织形态。结果烟熏组大鼠HCY水平较对照组升高(P<0.05);烟熏+戒烟组戒烟后HCY水平较烟熏组显著下降(P<0.05);烟熏+NAC组较烟熏组血浆HCY水平降低(P<0.05);烟熏+NAC+戒烟组HCY水平较烟熏组明显降低(P<0.05),且与对照组无显著差异(P>0.05)。组织病理学结果显示,烟熏组大鼠气管周围发现大片坏死组织,其间有大量炎性细胞浸润,组织出现明显的钙化改变。烟熏+NAC组、烟熏+戒烟组、烟熏+NAC+戒烟组的肺损伤程度均有不同程度改善。结论烟熏可导致大鼠肺部损伤,同时使血浆HCY水平升高;戒烟联合NAC可以减轻大鼠肺部损伤,显著降低血浆HCY水平。 Objective To investigate the effect of smoking cessation and N-acetylcysteine( NAC) intervention on rat lung tissue morphology and the plasma homocysteine( HCY) level. Methods A total of 50 male Wistar rats were divided into 5 groups such as the control group,the smoked group,the smoked + NAC intervention group,the smoked + smoking cessation group,and the smoked +NAC + smoking cessation group,with 10 rats in each group. Rats of 5 groups were blood sampled before experiment and 8 weeks intraexperiment,the rat venae angularis blood after centrifugation method was used to detect the ELISA level of HCY. HE staining was used to observe the morphological changes of lung tissue in rats. Results Compared with the control group,the HCY of smoked group increased( P〈0. 05);compared with the smoked group,the level of HCY in the smoked + smoking cessation group was significantly reduced( P〈0. 05),in smoked + NAC group reduced( P〈0. 05);in smoked + NAC + smoking cessation group was significantly reduced(P〈0. 05),and there was no significant difference with control group(P〈0. 05). Histopathological results showed that large necrotic tissue was surrounding trachea,there were a large number of inflammatory cells infiltration,tissue appeared obvious calcification changes. Smoked + NAC group,smoked + smoking cessation group,smoked + NAC + smoking cessation group can reduce lung injury in varying degrees. Conclusion Smoking cessation combined with NAC can significantly reduce HCY levels and reduce lung injury in varying degrees.
