摘要
目的以聚乳酸羟基乳酸(PLGA)为成膜材料,内载液态氟碳化合物(PFOB),外包裹金纳米壳层,构建金纳米壳液态氟碳聚乳酸纳米微囊,并进行其体外实验研究。方法运用单乳化水包油(O/W)溶剂挥发法制备包裹PFOB的纳米级PLGA粒子;以柠檬酸钠为还原剂,硼氢化钠为氧化剂制备金纳米粒子;使用静电吸附自组装法制备金纳米粒子聚乳酸微胶囊;最后采用表面原位引晶法制备金纳米壳聚乳酸微胶囊。结果金纳米粒子聚乳酸微胶囊粒子平均粒径为(268±80)nm,呈球形,分散性好;体外超声成像实验显示,金纳米壳聚乳酸微胶囊呈细腻均匀的点状密集高回声;细胞毒性试验显示材料无明显生物毒性,且体外溶液和细胞光热效果良好。结论成功制备集癌症成像与治疗为一体的金纳米壳液态氟碳聚乳酸微胶囊,具有良好的体外显像和光热治疗效果。
Objective To develop a contrast agent with PLGA as film forming and PFOB as internal material, which was covered by gold nanoshell. We will investigate its invitro photothermal effect and ultrasound imaging capability. Methods Using single emulsion oil-in-water(O/W) solvent evaporation method, the PFOB nanoseale PLGA particles was packaged; With sodium citrate as reducing angent, sodium borogydride as oxidizer as oxidizer, the gold nanoparticles were prepared; using electrostatic adsorption self-assembly, we prepared the gold nanopartieles polylactic acid microcapsule; finally, using in situ lead crystal surface, we prepared for gold nanoparticles shell polylactic acid microcapsule. Results The average particle size of gold nanoparicles polylactic acid microcapsule is (268± 80) nm, which is spherical, with good dispersion; The in vitro experiments demonstrate that the gold nanoshell polylactic acid microcapsule show exquisite and uniform dot intensive high echo with no obvious biological toxicity ; the in vitro solution and cell have good effect of photothermal. Conclusion This research successfully synthesizes the gold nanoparticles shell liquid fluorocarbon poly lactic-co-glycolic acid nano microcapsule. The effect of the gold nanoshell polylactic acid microcapsule in vitro imaging and photothermal therapy is good.
