HPLC法测定杏苏散提取物中活性成分苦杏仁苷和迷迭香酸含量

摘要 目的:采用反相高效液相色谱双波长法测定杏苏散水提液中苦杏仁苷和迷迭香酸的含量。方法:采用色谱柱为HPLC反向C18色谱柱(4.6mm×250mm,5μm)梯度洗脱,双波长为λ1=222nm,λ2=330nm,流速为1.0mL/min,柱温为30℃。结果:苦杏仁苷对照品线性关系良好,r=0.9999,回归方程为Y=27.390 X+0.0522,线性范围为0.08~0.64μg;迷迭香酸对照品线性关系良好,r=0.9997,回归方程为Y=26.141 X+0.0905,线性范围为0.04~0.32μg。苦杏仁苷平均加样回收率为98.59%,RSD=1.6%(n=6);苦杏仁苷平均加样回收率为98.86%,RSD=2.1%(n=6)。结论:方法简便、准确、重现性好,为杏苏散的质量控制和评价提供了一定的科学依据。 Objective:To explore the application of RP-HPLC in determining the contents of amygdalin and rosemary acid in XingSuSan extracts.Methods:The gradient elution with HPLC reversed chromatographic column C18(4.6mm×250mm,5μm)had been used with the dual wavelengths respectivelyλ1=222nm andλ2=330nm,the flow rate 1.0mL/min,and the column temperature at 30℃.Results:The calibration curve of amygdalin was linear in the range of 0.08~0.64μg,r=0.9999;the linear equation was Y=27.390 X+0.0522.The calibration curve of rosemary acid was linear in the range of 0.04~0.32μg,r=0.9997;the linear equation was Y=26.141 X+0.0905.The average recovery of amygdalin was 98.59%,RSD=1.6%(n=6),the average recovery of rosemary acid 98.86%,RSD=2.1%(n=6).Conclusion:The HPLC method is simple,accurate and reproducible,which provides a scientific foundation for XingSuSan quality control and evaluation.
