电针三阴交对AD小鼠学习记忆能力及海马区Ach、AchE、ChAT的影响 被引量:21

摘要 目的观察电针"三阴交穴"对SAMP8快速衰老模型小鼠学习记忆能力和脑组织海马区内乙酰胆碱(Ach)、乙酰胆碱酯酶(Ach E)、乙酰胆碱转移酶(Ch AT)的影响。方法将快速老化SAMP8小鼠随机分为模型组和电针组,并设同源SAMR1小鼠作为对照组,每组均为6只。电针组给予三阴交电针干预;对照组,模型组常规抓取,不进行针刺。采用Morris水迷宫法检测小鼠的学习记忆能力;酶联免疫法(ELISA)检测小鼠海马部位的乙酰胆碱含量、乙酰胆碱酯酶和乙酰胆碱转移酶活性。结果 Morris水迷宫定位航行实验和空间探索实验显示:较SAMR1小鼠相比SAMP8小鼠学习记忆能力显著下降(P<0.01),电针治疗后明显改善(P<0.05),差异有统计学意义。指标显示:模型组较对照组海马内AchE、ChAT活性明显降低,Ach含量显著减少(P<0.01),电针治疗后AchE、ChAT活性明显升高,且Ach含量明显增多(P<0.05),差异有统计学意义。结论电针三阴交穴可以明显改善老年痴呆模型小鼠学习记忆能力并提高海马区乙酰胆碱含量,其作用可能是通过改善脑组织海马区乙酰胆碱酯酶和乙酰胆碱转移酶活性来实现的。 Objective To observe the effects of electroactupuncture (EA) at "Sanyinjiao" on learning and memory abilities and brain Ach, AchE and ChAT activities in a SAMP8 mouse model of senile dementia. Mothods The SAMP8 mice were randomly divided into two groups: model group and EA group. The homologous SAMR1 was used as control group. Each team has six mice. EA group was applied to bilateral "sanyinjiao"; Control group and model group were given constraint and no EA treatment. Morris water maze tests were coculated for the SAMP8 mice's learning and memory ability. The hippocampus tissues were collected for detecting the contents of Ach and the activity of AchE and ChAT by ELISA. Results Morris water maze tests results, containing the navigation tests and the space exploration experiments: Compared with the SAMR1 mice, the learning and memory ability of SAMP8 mice were significantly declined (P〈0.01). After the treatment with EA, the ability were significantly improved(P〈0.05), and the differences was statically significant.Detection index results: compared with the control group, the Ach content and the activity of AchE and ChAT in hippocampus of model group were lower(P〈0.01), while after the treatment the level were significantly improved(P〈0.05), and the differences was statically significant. Conclusion EA of "Sanyinjiao" can obviously improved the learning and memory ability in AD mice and improved the Ach content in hippocampus. The effect may be achieved by improving the AchE and ChAT activity of hippocampus in AD mice.
