摘要
目的:研究丹参酮Ⅰ对肝癌细胞HepG2生长的影响。方法:MTT法检测丹参酮Ⅰ对HepG2细胞增殖的影响,Western印迹检测经丹参酮Ⅰ处理的HepG2细胞中凋亡标志物c-Parp和周期蛋白D1的表达变化,流式细胞术检测细胞的凋亡及周期分布。结果:MTT分析发现丹参酮Ⅰ可抑制HepG2细胞的生长;经丹参酮Ⅰ处理的HepG2细胞中凋亡标志物c-Parp表达升高,周期蛋白D1表达无明显变化;流式细胞术分析结果进一步证明丹参酮Ⅰ可诱导HepG2细胞发生凋亡,对细胞周期分布无明显影响。结论:丹参酮Ⅰ通过诱导细胞凋亡而抑制HepG2细胞的生长,对细胞周期无明显阻滞,为进一步研究丹参酮Ⅰ的抗肿瘤作用分子机制提供了线索。
Objective:To study the influence of TanshinoneⅠon HepG2 cells proliferation,cell cycle and cell apoptosis. Methods:The proliferation of cells was detected by MTT assay. Western blotting was used to detect the c-Parp and cyclin D1. The cell apoptosis and cell cycle were detected by flow cytometer. Results:MTT results showed that tanshinoneⅠinhibits the growth of HepG2 cells. Western blotting proved that tanshinoneⅠcan make the apoptosis marker c-Parp increase in HepG2 cells,but didn't change cyclin D1. Flow cytometry results showed that tanshinoneⅠcan induce apoptosis of HepG2 cells and had no effect on the cycle distribution. Conclusion:TanshinoneⅠ inhibits the growth of HepG2 cells by inducing apoptosis,which provided a clue for further study on the molecular mechanism of anti-tumor action of tanshinoneⅠ.
