摘要
目的:研究胰腺癌组织中GSK3β表达情况与细胞侵袭特性、增殖特性的相关性。方法:选择2013年5月~2016年3月期间在湖北省武汉市新洲区中医医院手术切除的胰腺癌组织和癌旁组织,采用免疫组化试剂盒测定GSK3β的阳性表达率,采用酶联免疫吸附试剂盒测定GSK3β以及侵袭相关分子、增殖相关分子的蛋白表达量。结果:胰腺癌组织中GSK3β的阳性表达率以及蛋白含量均显著高于癌旁组织;胰腺癌组织中CCL21、CCR7、Vimentin、Cyclin D1、CDK4、CDK6的表达量显著高于癌旁组织,E-cadherin、P15INK4B、P21WAF1/CIP1的表达量显著低于癌旁组织;GSK3β阳性表达胰腺癌组织中CCL21、CCR7、Vimentin、Cyclin D1、CDK4、CDK6的表达量显著高于GSK3β阴性表达胰腺癌组织,E-cadherin、P15INK4B、P21WAF1/CIP1的表达量显著低于GSK3β阴性表达胰腺癌组织。结论:胰腺癌组织中高表达的GSK3β对癌细胞的侵袭、增殖具有促进作用。
Objective:To study the correlation of GSK3βexpression with cell invasion and proliferation in pancreatic cancer tissue.Methods:Pancreatic cancer tissue and para-carcinoma tissue surgically removed in our hospital between May 2013 and March 2016 were selected,immunohistochemical kits were used to determine the positive expression rate of GSK3β,and enzyme-linked immunosorbent assay kits were used to determine the protein expression of GSK3βas well as invasion and proliferation-related molecules.Results:The positive expression rate and protein level of GSK3 in pancreatic cancer tissue were significantly higher than those in para-carcinoma tissue;CCL21,CCR7,Vimentin,Cyclin D1,CDK4 and CDK6expression in pancreatic cancer tissue were significantly higher than those in para-carcinoma tissue while E-cadherin,P15INK4 B and P21WAF1/CIP1 expression were significantly lower than those in para-carcinoma tissue;CCL21,CCR7,Vimentin,Cyclin D1,CDK4 and CDK6expression in pancreatic cancer tissue with positive GSK3βexpression were significantly higher than those in pancreatic cancer tissue with negative GSK3βexpression while E-cadherin,P15INK4 Band P21WAF1/CIP1 expression were significantly lower than those in pancreatic cancer tissue with negative GSK3βexpression.Conclusions:Highly expressed GSK3βin pancreatic cancer tissue can promote cancer cell invasion and proliferation.
