摘要
为了研究CFL2基因与骨骼肌纤维相关基因之间的关系,本试验运用已筛选的CFL2低表达细胞株,利用实时定量和Western blot技术检测CFL2基因低表达对肌纤维组成成分MLC、α-actin与肌纤维类型MyHCslow、MyHC2b、MyHC2a、MyHC2x表达的影响。结果表明,低表达CFL2后,肌纤维组成成分MLC和α-actin的表达均显著上调(P<0.05)。MyHC2x、MyHC2b、MyHC2a、MyHCslow的表达均显著下调(P<0.05)。说明CFL2可能起到通过调节MyHC类型转换来达到调节红白肌肉比例的效果,进而直接影响猪肉品质。
The present study was conducted to examine the relationship between CFL2 gene and skeletal muscle fiber.The expressions muscle fiber(MLC,α-actin) and myosin heavy chain(MyHC2a,MyHC2b,MyHC2x and MyHCslow) related genes were investigated by realtime PCR and western blot in CFL2 low-expressed cells.The results indicated that expressions of MLC and α-actin were both significantly increased(P0.05); and expressions of MyHC2x,MyHC2b and MyHC2a,MyHCs were significantly decreased(P 0.05).The results suggested that CFL2 might regulate the proportion of red muscle and white muscle by converting the MyHC type,which might directly affect the meat quality.
