摘要
目的:研究消癌平联合5-氟尿嘧啶处理对肝癌细胞体外生长及癌基因表达的影响。方法:培养肝癌细胞株SMMC-7721并用不同剂量消癌平(10、20、40μL/mL)及5-氟尿嘧啶(10、20、40μmol/mL)处理。处理后12h、24h、48h时,采用CCK-8试剂盒测定细胞活力;处理后24h时,采用酶联免疫吸附试剂盒测定增殖、凋亡基因的表达。结果:处理后12、24、48h时,不同剂量消癌平和5-氟尿嘧啶均能以剂量依赖性的方式降低细胞增殖活力且40μL/mL消癌平+40μmol/mL 5-Fu组的细胞增殖活力均显著低于40μL/mL消癌平组、40μmol/mL 5-Fu组;处理后24h时,40μL/mL消癌平+40μmol/ml5-Fu组、40μL/mL消癌平组、40μmol/mL 5-Fu组细胞中P16^(INK4)、P57^(kip2)、Caspase-3、PDCD5的蛋白表达量均显著高于对照组,LETM1、SIRT6、UHRF1的蛋白表达量均显著低于显著低于对照组且40μL/mL消癌平+40μmol/mL 5-Fu组细胞中P16^(INK4)、P57^(kip2)、Caspase-3、PDCD5的蛋白表达量均显著高于40μL/mL消癌平组、40μmol/mL 5-Fu组,LETM1、SIRT6、UHRF1的蛋白表达量均显著低于40μL/mL消癌平组、40μmol/ml 5-Fu组。结论:消癌平联合5-氟尿嘧啶对肝癌细胞的增殖具有抑制作用,同时能够增加凋亡基因的表达、降低增殖基因的表达。
Objective: To study the effect of Xiaoaiping combined with 5-fluorouracil treatment on liver cancer cell growth in vitro and oncogene expression. Methods: Liver cancer cell lines SMMC-7721 were cultured and treated with different doses of Xiaoaiping (10, 20 and 40/A/mL) and 5-fluorouracil (10, 20 and 40 μmol/mL). 12h, 24h and 48h after treatment, the CCK-8 kits were used to determine the cell viability; 24h after treatment, enzyme-linked immunosorbent assay kits were used to determine proliferation and apoptosis gene expression. Results: 12h, 24h and 48h after treatment, different doses of Xi- aoaiping and 5-fluorouracil could all reduce cell proliferation activity in dose-dependent manner and the cell proliferation activity of 40 μl/mL Xiaoaiping -P 40 μmol/mL 5-Fu group were significantly lower than those of 40 μl/mL Xiaoaiping group and 40 /Lmol/mL 5-Fu group; 24h after treatment, P16INK4 , P57^kip2 , Caspase-3 and PDCD5 protein expression in 40 btL/mL Xiaoaip- ing q- 40/~mol/mL 5-Fu group, 40 pL/mL Xiaoaiping group and 40μmol/mL 5-Fu group were significantly higher than those in control group while LETM1, SIRT6 and UHRF1 protein expression were significantly lower than those in control group, and P16^INK4 , P57kip2 , Caspase-3 and PDCD5 protein expression in 40 μL/mL Xiaoaiping + 40μmot/ml 5-Fu group were sig- nificantly higher than those in 40 μL/mL Xiaoaiping group and 40 /μmol/mL 5-Fu group while LETM1, SIRT6 and UHRF1 protein expression were significantly lower than those in 40 μl/mL Xiaoaiping group and 40μmol/ml 5-Fu group. Conclusion.. Xiaoaiping combined with 5-fluorouraeil has inhibitory effect on liver cancer cell proliferation, and can also increase the expres- sion of apoptosis genes and reduce the expression of proliferation genes.
