摘要
Doublecortin样激酶-1(doublecortin-like kinase 1,DCLK1)最初是在神经系统中发现的一种跨膜微管相关蛋白激酶,具有调节微管聚合以及促进神经元迁移的功能。近年来,随着DCLK1在消化道恶性肿瘤中的研究,其被认为是一种在恶性肿瘤形成和发展中发挥重要作用的蛋白质。此外,DCLK1可以通过调控多种mi RNAs促进上皮间质转化,进而促进肿瘤的侵袭和转移。研究DCLK1在恶性肿瘤中发挥的作用及机制并特异性阻断其分子机制,或许可以成为一种治疗肿瘤的新方法。
Doublecortin-like kinase 1 (DCLK1), initially described in neural system, is a microtubule-associated protein that plays key role in the regulation of neural cell differentiation, migration, and apoptosis. Recently, accumulating evidence indicates that DCLK1 may be an essential factor in initiation and progression of gastrointestinal tumors. Furthermore, this kinase induces epithelial-mesenchymal transition (EMT) and thus promotes tumor metastasis via microRNA-dependent pathways. Investigating the biological role of DCLK1 in tumors and specifically blocking DCLKl-dependent pathways might be a promising therapeutic target for treatment of gastrointestinal tract tumors. This review highlights new findings regarding the role of DCLK1 in tumor biology.
