摘要
研究了不同萃取方式对山丁子多酚得率及抗氧化活性的影响。采用微波萃取、铜极板和钢极板等离子萃取、聚能式和清洗式超声波萃取及真空萃取等方法提取山丁子中的抗氧化物质。通过体外抗氧化能力分析可知,山丁子多酚有较好的抗氧化能力,微波萃取效果最佳。以多酚得率、总抗氧化能力和DPPH清除率为指标,采用响应面法优化了山丁子多酚提取工艺并得出了最佳工艺条件:山丁子多酚的提取工艺为料液比1∶30(g/m L)、微波功率572 W、处理时间21 min,此条件下多酚提取率为4.211 mg/g;山丁子总抗氧化物质的提取工艺为料液比1∶32(g/m L)、微波功率689 W、处理时间20 min,此条件下总抗氧化能力为11.12 U/m L;山丁子DPPH清除率的提取工艺为料液比1∶33(g/m L)、微波功率697 W、处理时间20 min,此条件下DPPH清除率为80.02%。
The effects of different extraction methods on polyphenols yield and antioxidant activity of Malus baccata were studied. Microwave extraction, copper plate and steel plate plasma extraction, energy-gathered and cleaning ultrasonic extraction, and vacuum extraction were used to extract the antioxidant substances in the Malus baccata. Analysis of antioxidant capacity in vitro showed that Malus baccata had excellent antioxidant capacity and microwave was the best extraction method. Using the polyphenol yield, total antioxidant capacity and the DPPH clearance rate as index, the optimization extraction conditions of polyphenols were obtained as follows through response surface methodology: extraction condition of polyphenols was solid-liquid ratio 1 : 30 ( g/mL ) , microwave power 572 W and processing time 21 min, resulting in the polyphenols extraction yield of 4.211 mg/g; extraction condition of total antioxidant was solid-liquid ratio 1 : 32 ( g/mL ) , microwave power 689 W, processing time 20 min, resulting in the total antioxidant capacity of 11.12 U/mL; extraction condition of DPPH clearance rate was solid-liquid ratio 1 : 33 ( g/mL ), microwave power 697 W and processing time 20 min, resulting in the DPPH clearancerate of 80.02%.
