摘要
建立高效液相色谱测定中药丹参中丹参酮Ⅰ,隐丹参酮,丹参酮ⅡA含量的方法。方法采用C_(18)色谱柱(4.6 mm×250 mm,5μm),以乙腈和0.2%磷酸水溶液为流动相,采用梯度洗脱,流速为1.0 mL/min;检测波长为254 nm;柱温30℃。丹参酮Ⅰ,隐丹参酮,丹参酮ⅡA三种化合物在1.56×10^(-6)5×10^(-5)g/mL(r>0.999)浓度范围内线性关系良好。该方法快速、灵敏、便于操作且可靠性高,适用于中药样品中丹参酮类化合物的测定。
To establish a HPLC method for the determination of Tanshinone I, Cryptotanshinone and Tanshinone IIA in Radix Salviae Mihiorrhizae, the HPLC was performed on a ZORBAX SB-C18 column (4.6 × 250 mm, 5 μm) by a gradient elution using 0. 2% H3PO4 and acetonitrile as the mobile phase. The flow rate was 1.0 mL/min, the detection wavelength was set at 254 nm and column temperature was 30℃. The results showed good linearity in the range of 1.56× 10-6-1--5× 10-5 g/mL (r〉0. 999)for Tanshinone I, Cryptotanshinone and Tanshinone IIA. The method was sensitive, accurate and reliable for the determination of the three components in Radix Salviae Mihiorrhizae.
