摘要
目的:探讨mTOR信号通路对非小细胞肺癌放疗敏感性的影响。方法:将非小细胞肺癌细胞系A549细胞分成对照组与实验组,对照组不对mTOR信号通路进行干预,实验组采用不同剂量的雷帕霉素干预mTOR信号通路,两组采用不同剂量射线辐射,对比分析两组细胞的存活能力、克隆数目、凋亡情况。结果:实验组细胞的平均存活时间为(3.5±0.7)d,对照组细胞的平均存活时间为(5.8±0.6)d,实验组明显短于对照组,对比两组差异显著,有统计学意义(P<0.05);实验组细胞4Gy、8Gy的平均克隆数目分别为(55±6)个、(19±2)个;对照组细胞4Gy、8Gy的平均克隆数目分别为(144±21)个、(96±13)个,实验组明显低于对照组,对比两组差异显著,均有统计学意义(P<0.05);实验组细胞的凋亡比例明显高于对照组,对比两组差异显著,有统计学意义(P<0.05)。结论:mTOR信号通路对非小细胞肺癌放疗敏感性具有一定的影响,抑制mTOR信号通路可增强非小细胞肺癌放疗的敏感性。
Objective: To explore the effect ofmTOR signaling pathway on radiosensitivity of non-small cell lung cancer. Methods: Non small cell lung cancer cell line A549 Cells were divided into the control group and the experimental group, the control group received no mTOR pathway intervention while the experimental group received different doses of rapamycin to intervene mTOR signaling pathway, two groups received different doses of radiation, a comparative analysis of two groups of cell viability, clone number and apoptosis was made. Results: The mean survival time of the experimental group and the control group was (3.5 ± 0.7) d and (5.8 ± 0.6) d respectively, the mean survival time of the experimental group was significantly shorter than that of the control group, the difference was statistically significant (P〈0.05); The average number of clones 4Gy and 8Gy in the experimental group was (55 ±6) and (19 ± 2) while that of the control group was (144 ± 21) and (96 ± 13), the number of the experimental group was significantly less than that of the control group, the difference was statistically significant (P〈0.05); The apoptosis rate of the experimental group was significantly higher than that of.the control group, the difference was statistically significant (P〈0.05). Conclusion: MTOR signaling pathway had a certain effect on the radiosensitivity of non-small cell lung cancer, inhibition of mTOR signaling pathway could enhance the radiosensitivity of non-small cell lung cancer.
