摘要
目的:探讨二十五味儿茶丸对尿酸钠诱导的大鼠急性痛风性关节炎的防治作用及炎症反应机制。方法:80只SD大鼠,随机分为正常组,模型组,秋水仙碱组(0.3 mg·kg^(-1)),氢化可的松组(4 mg·kg^(-1))及二十五味儿茶丸高、中、低剂量组(2.4,1.2,0.6 g·kg^(-1)),除正常组外,其余各组采用向大鼠踝关节内注入尿酸晶体建立急性痛风性关节炎模型,测定造模前及造模后3,6,9,12,15 h关节肿胀度,同时经眼眶静脉丛取血测定白细胞(WBC)和中性粒细胞(NEUT)数目,取滑膜组织测定炎性因子肿瘤坏死因子-α(TNF-α),白细胞介素-1β(IL-1β),IL-8,环氧合酶-2(COX-2)含量,并采用蛋白质免疫印迹(Western bolt)检测滑膜组织中COX-2蛋白水平的表达。结果:与正常组比较,模型组各时间点大鼠关节肿胀度明显升高,WBC,NEUT数目明显升高,TNF-α,IL-1β,IL-8,COX-2含量明显升高(P<0.05,P<0.01),COX-2蛋白水平有所升高,但无统计学差异;与模型组比较,二十五味儿茶丸高、中、低剂量组肿胀度明显降低,抑制率明显升高,可明显减少WBC,NEUT数目,明显降低炎性因子TNF-α,IL-1β,IL-8,COX-2含量,并可明显降低COX-2蛋白水平表达(P<0.05,P<0.01)。结论:二十五味儿茶丸通过减少白细胞、中性粒细胞数目,抑制炎症因子的释放,从而达到抗急性痛风性关节炎炎症反应作用。
Objective: To investigate the effect of Ershiwuwei Ercha pill in acute gouty arthritis rats induced by sodium urate crystals. Method: A total of 80 SD rats were randomly divided into blank group,model group,colchicine group( 0. 3 mg·kg^-1),hydrocortisone group( 4 mg·kg^-1) and twenty-five taste tea pill different dose group( 2. 4,1. 2,0. 6 g·kg^-1). Except for blank group,all of the remaining groups were included into the acute gout arthritis model by injecting uric acid into the rat ankle joint. The joint swelling degree was measured after making model at 0,3,6,9,12,15. At the same time,white blood cell( WBC) and neutrophil( NEUT)were determined by analyzingblood cell from orbital venous plexus,and the levels of tumour necrosis factor-α( TNF-α),interleukin^-1β( IL^-1β),IL-8,cyclooxygenase-2( COX-2) in synovial tissues were determined by ELISA,and the expression of COX-2 protein in synovial tissues was detected by Western blot. Result: Compared with the blank group,the joint swelling degree of rats in the model group was significantly increased at each time point. The numbers of WBC and NEUT were significantly increased,the level of TNF-α,IL^-1β,IL-8,COX-2were significantly increased( P〈0. 05,P〈0. 01),the expression of COX-2 protein were slightly increased,but with no statistical difference. Compared with the model,the joint swelling degree of rats in the high,middle and low-dose Ershiwuwei Ercha pill groups were significantly reduced,with a significant increase in inhibition ratio.The numbers of WBC and NEUT were significantly reduced. The level of TNF-α,IL^-1β,IL-8, COX-2 were significantly decreased. The expression of COX-2 protein were significantly reduced( P〈0. 05,P〈0. 01).Conclusion: Ershiwuwei Ercha pill can reduce joint swelling degree, leukocyte,neutrophil cell number and inflammatory factors release to resist inflammatory reaction.
