摘要 目的探讨手术切除的肺癌组织块移植于裸鼠皮下建立人肺癌裸鼠移植瘤模型的方法可操作性及移植瘤的生物学特性.方法将术中切除4例非小细胞肺癌标本无菌条件下移植于裸鼠皮下,观察肿瘤生长情况,记录生长曲线.通过常规病理学检查、免疫组化分析及荧光定量PCR分析,将成瘤的移植瘤与原肿瘤进行对比,观察其生物学特性变化.结果 4例肺癌标本中建模成功2例,成瘤率为50%.病理检查及免疫组化分析结果提示移植瘤生物学特性与原肿瘤保持较高的相似性,荧光定量PCR检测结果提示移植瘤所表达的基因为人源基因.基因及蛋白表达证实建模成功,移植瘤为人源性肿瘤.结论组织块法皮下移植建立人非小细胞肺癌裸鼠移植瘤模型是一种高效可行的方法,成瘤率高,所建模型能最大程度模拟原肿瘤的生物学特性. Objective To study the way of establishing patient-derived lung cancer xenograft models and discovering the biological characteristics of xenografts. Me thods NSCLC tissue were collected from 4 primary lung cancer patients and implanted in subcutaneous layer of the nude mice. The growth condition and groph of tumors were observed and recorded,and the histopathology,immunohistochemistry and real-time PCR were used to confirm the graft tumors were or not derived from the patients. Re s ults Two out of four xenograft models were successfully established. HE staining and immunohistochemistry analysis confirmed that major histological features of the original tumors were retained in xenografts. The expression of Ki67 and mitochondria were evaluated by q PCR, results revealed that xenografts had a tight correlation with primary patient tumors. Conclus ions The subcutaneous layer xenograft approach was a feasible way to establish xenograft models derived from patient tumor. The xenografts recapitulate the genotype and phenotype of original patient tumors.
