摘要 以食品安全国家标准《食品中乳铁蛋白的测定(征求意见稿)》为参考,优化预处理步骤和梯度洗脱条件,肝素亲和柱净化,紫外高效液相色谱法测定婴幼儿配方羊奶粉中乳铁蛋白含量。检测波长280 nm,乳铁蛋白标准品出峰时间9.475min。乳铁蛋白标准品工作液浓度50~250μg/mL,在此范围内线性回归方程y=129.67x-7.4719,R^2>0.9991。乳铁蛋白定性重复性RSD%(n=6)为0.074,定量重复性RSD%(n=6)为0.996。乳铁蛋白加标回收率在93.4%~99.1%,平均回收率96.7%。 The determination of lactoferrin in food ( draft for comments ) was taken as the reference in the national food safety standard. The pretreatment and gradient elution conditions were optimized. The heparin affinity column was used for the determination of lactoferrin in infant formula sheep powder. Detection wavelength was 280 nm, lactoferrin standard peak time was 9.475 min. Lactoferrin standard working solution concentration was 50- 250 μg / mL, in this range linear regression equation was y=129.67x-7.4719, R2〉 0.9991. The qualitative repeatability RSD% ( n =6 ) of lactoferrin was 0.074 and the quantitative repeatability RSD% ( n=6 ) was 0.996. The recoveries of lactoferrin were 93.4% - 99.1%, and the average recovery was 96.7%.
