结核患者外周血αβT和γδT细胞亚群免疫亚型细胞分布的探讨 被引量:4

摘要 目的:检测健康成人和结核患者外周血αβT细胞亚群以及γδT细胞亚群分布情况。方法:采集健康成人及结核病患者外周血,用不同荧光素标记单抗染色后,用流式细胞术检测αβT细胞免疫亚型CD45RA+CD27+初始型细胞(Tnaive)、CD45RA-CD27中央记忆型T细胞(Tcm)+、CD45RA-CD27-效应记忆型T细胞(Tem)和CD45RA+CD27-效应型T细胞(Teff);γδT细胞中Vδ1、Vδ2、Vδ1-Vδ2-(即Vδ3-8)4种免疫亚型的百分率。结果:与健康成人比较,结核患者CD4+αβT细胞中Tnaive、Teff细胞的百分率降低(P<0.01),Tem细胞的百分率升高(P<0.01),Tcm细胞差异无统计学意义(P>0.05)。结核与健康成人比较,患者CD8+αβT细胞中Tnaive、Tcm细胞的百分率降低(P<0.01和P<0.05),Tem、Teff细胞的百分率升高(P<0.01)。与健康成人比较,结核患者Vδ1γδT细胞中Tnaive、Tcm细胞的百分率降低(P<0.05),Tem细胞的百分率升高(P<0.05),Teff细胞差异无统计学意义(P>0.05);结核患者Vδ2γδT细胞中Tcm细胞的百分率降低(P<0.05),Tem、Teff细胞的百分率升高(P<0.05,P<0.01),Tnaive细胞差异无统计学意义(P>0.05);结核患者Vδ3-8γδT细胞中Tem细胞的百分率升高(P<0.05),Tnaive、Tcm、Teff细胞差异无统计学意义(P>0.05)。结论:结核病患者外周血中αβT和γδT细胞亚群分布均有显著改变。 Objective :To investigate the distribution of immune subtpyes of αβ and ~ST cell subsets of peripheral blood in healthy adults and pulmonary tuberculosis (TB) patients. Methods:The peripheral blood from healthy adults and TB patients were stained with different fluorochrome-conjugated mAbs. The CIM5RA+ CD27+ naive T cells (Tnaive), CD45RA-CD27+ central memory T cells (Tcm) ,CD45RA-CD27- effector memory T cells(Tem) and CIM5RA+ CD27- effector T cells of αβ T cells immune subtpyes,and the proportions of the Vδ1, Vδ2, Vδ1 - Vδ2 - ( V83-8 ) immune subtypes T cells in γδ T cells were detected using flow cytometry. Results: Compared with healthy adults, the percentages of Tnaive & Teff cells, and Tem ceils in CD4+ αβ T cells of TB patients decreased and increased, respectively (P 〈 0.01 ), and the difference of Tcm ceil between healthy adults and TB patients was not statistically significant ( P 〉 0.05 ). Compared with healthy adults, the percentages of Tnaive & Tcm ceils, and Tem &Teff cells decreased and increased in CD8 + αβ T ceils of TB patients, respectively(P 〈 0.01 and P 〈 0.05 ). Compared with healthy adults, the percentages of Tnaive & Tcm cells, and Tem ceils decreased and increased in CD8 + αβ T cells of TB patients, respectively ( P 〈 0.05 ), and the difference of Teff cells between healthy adults and TB patients was not statistically significant( P 〉 0.05 ). The percentages of Tcm cells, and Tem& Teff cells in Vδ2 γδ T cells of rib patients decreased and increased, respectively(P 〈 0.05 and P 〈 0.01 ), and the difference of Tnaive ceils between healthy adults and TB patients was not statistically significant( P 〉 0.05 ). The percentage of Tem ceils in Vδ3-8 γδ T cells of tuberculosis patients increased( P 〈 0.05 ), and the differences of the percentages of Tnaive, Tcm and Teffcells between healthy adults and TB patients were not statistically significant(P 〉 0.05). Conclusions:The changes of the αβ T and γδ T cell subsets distributions in peripheral blood of TB patients are significant.
