肾穿六胺银套染马松三色双重染色的改进

Modified Double Stain of PASM and Masson Trichrom for Renal Needle Biopsy

目的探讨改进肾穿六胺银套染马松三色的染液与染色方法的可行性。方法收集2015年1月—2016年12月厦门大学附属第一医院病理科已确诊的100例肾穿刺活检病例的蜡块,每例蜡块连续切片2片,随机分为两组。甲组采用传统染液与传统染色法染色,乙组采用改进染液与改进染色法染色,以百分制评定200张六胺银套染马松三色染色片的得分,统计各组的得分,结果用(x±s)表示,采用独立样本t检验进行比较,分析两组得分的差异是否有统计学意义,并比较两组显微镜下的染色结果。结果乙组染色的得分高于甲组[(88.7±9.6)分vs.(72.2±10.3)分],差异有统计学意义(t=11.719,P<0.001);乙组切片镜下显示肾小球鲍曼囊和毛细血管襻基膜均呈黑色细丝状,细胞核呈紫蓝色,网状纤维和弹力纤维呈黑色,免疫复合物、细胞质与红细胞均呈红色,细胞间质与胶原纤维呈绿色,组织结构清晰,着色均匀,优于甲组。结论改进染液与染色方法使六胺银套染马松三色的染色效果好。

Objective To explore the effect of modified double stain of PASM and Masson trichrom for renal needle biopsy dyeing dye solutioo preparation. Methods To collecte 100 cases of renal biopsy diagnosis of wax in the first affiliated hospital of xiamen university from January 2015 to December 2016, each wax of 100 cases was cut into two serial sections and divided into two groups randomly. Group A division was dyed by traditional dye solution under the traditional dyeing method. Group B division was dyed with modified method. Evaluation of 200 six silver amine or Masson staining score sheet. The scores of each group were presented in the form of （ x ＋ s ） . To with the independent sample t-test, and analyze the statistical significance, and the staining results of were compared between the two groups under the microscope. Results The results of group B staining score higher than that of group A [（88.7±9.6） vs. （72.2±10.3）], the difference was statistically significant （t = 11.719, P 〈 0.001）. Under the microscope, the result of group B slices showed that bauman capsule and glomerular capillaryloop in basement membrane were black filaments, purple and blue nuclei, reticular fibers and elastic fibers were black, the immune complexes, cytoplasm and red blood cells were red, the intercellular and collagen fibers were green, clear boundaries and uniform coloring, better than group A. Conclusion The modified double stain of PASM and Masson trichrom dye solution preparation method and improved dyeing method is significantly better than the traditional method.
