DNA甲基化转移酶在急性A型主动脉夹层发病机制中的作用

The Involvement of DNA Methylation Transferases in the Pathogenesis of Type A Acute Aortic Dissection

摘要 目的探讨DNA甲基化转移酶(DNMTs)与急性A型主动脉夹层(Acute aortic dissection,AAD)发生发展之间的关系。方法收集5例急性A型AAD患者病变血管组织和4例无主动脉疾病的器官捐献者的升主动脉血管组织,采用Western Blot方法检测DNMTs(DNMT1、DNMT3A、DNMT3B、DNMT3L)的蛋白在患者和正常对照血管组织中的表达情况。结果与健康对照组相比,整体上DNA甲基化转移酶蛋白在A型AAD患者病变血管组织中表达下降,DNMT1蛋白在A型AAD患者病变血管组织中表达较正常对照下降约50%(P=0.023),DNMT3B蛋白在A型AAD患者病变血管组织中表达下降约50%(P=0.009);DNMT3A和DNMT3L蛋白在A型AAD患者病变血管组织中表达下降趋势,尽管无统计学意义(P=0.414,P=0.16)。结论 DNA甲基化转移酶在急性A型AAD患者病变血管组织中表达水平较正常对照呈下降趋势,DNA甲基化是否参与A型AAD病变的发生发展尚需进一步研究。 Objective To determine the role of DNA methylation transferase in type A actue aortic dissection (AAD). Method Ascending aortic tissue samples were collected from 5 cases of type A AAD and 4 eases of organ donors without aortic diseases. Western Blot was performed to demonstrate the protein expression of four members of DNA methylation transferases in the ascending aortic tissue. Results About 50% decrease was observed in the protein expression of DNMTl(P=0.023) and DNMT3B(P=0.009) as type A AAD compared with healthy controls. Both the protein expression of DNMT3A and DNMT3L showed a decreased trend although the differences were not statistically significant(P=0.414 and P=0.16, respectively). Conclusions Our results indicate abnormal expression of proteins of DNA methylation trasferasesin aortic tissue from type A AAD. The role of DNA methylationin the development and process of AAD need to be investigated in future study.
