不同牙周状态下正畸力诱导Th1和Th2细胞的表达及其比率变化的初步研究 被引量:8

摘要 目的研究不同牙周状态下正畸力诱导外周血Th1细胞和Th2细胞的表达及Th1/Th2细胞比率的变化,初步探讨牙周炎正畸治疗机制。方法 14只8周龄SD大鼠随机分为牙周炎静止期移动组(RM组)、正常牙周移动组(CM组)、牙周炎活动期对照组(A组)、牙周炎静止期对照组(R组)及空白对照组(NC组)。采用丝线结扎并局部注射脂多糖法建立牙周炎活动期模型,建模成功后去除上述刺激因素并行牙周治疗,建立牙周炎静止期模型。各移动组大鼠用镍钛拉簧以50g力近中移动上颌第一磨牙,于移动后第7天处死。流式细胞技术检测大鼠外周血Th1细胞及Th2细胞的表达及Th1/Th2比率变化。结果 RM组和CM组、A组、R组外周血Th1细胞、Th2细胞比例及Th1/Th2比率均较NC组升高(P<0.05)。与CM组相比,RM组外周血Th1细胞、Th2细胞比例升高(P<0.05),Th1/Th2细胞比率降低(P<0.05);与A组(P<0.05)相比,RM组外周血Th1细胞比例降低(P<0.05),Th2细胞比例升高(P<0.05),Th1/Th2细胞比率降低(P<0.05)。结论 Th1、Th2细胞参与调节正常牙周及牙周炎静止期正畸牙齿移动。相对于正常牙周,牙周炎静止期正畸力诱导Th1、Th2细胞在牙周组织的表达增强,以Th2细胞为显著,Th1/Th2比率减小。 Objective To examine the expression of Thl and Th2 cells and the change of Thl/Th2 ratio in peripheral blood during orthodontic tooth movement under different periodontal conditions. Methods Fourteen 8-week-old SD rats were randomly divided into recovery periodontitis movement group (RM) and normal periodontal movement group (CM) and active periodontitis group (A) and recovery periodontitis control group (R) and the normal control group (NC). Active periodontitis model was established by silk thread ligation and local injection of lipopolysaecharide. These stimulating factors were removed when active periodontitis was induced, and then recovery periodontitis model was set up. NiTi spring was used to apply 50g mesial force to move upper first molar of RM and CM groups. RM and CM rats were sacrificed on day 7. The expression of Thl cell and Th2 cell and the change of Thl/Th2 ratio in peripheral blood were detected by flow cytometry. Results Thl and Th2 cells and Thl/Th2 ratio in groups RM, CM, A and R were increased compared to the group NC ( P 〈 0.05 ). Thl and Th2 cells were increased and Thl/Th2 ratio was decreased in group RM compared to group CM (P 〈0.05 ). Th2 cells were increased, Thl cells and Thl/Th2 ratio were decreased in group RM compared to the group A ( P 〈 0.05 ). Condusion Thl and Th2 cells participate in the normal periodontal and recovery periodontitis orthodontic tooth movement. The expression of Thl and Th2 in the recovery periodontitis was up-regulated, with Th2 cells more significantly, and Thl/Th2 ratio was decreased.
