摘要
目的应用99Tcm-TRODAT-1 SPECT/CT显像评估7, 8-二羟基黄酮(7,8-DHF)对正常食蟹猴纹状体(ST)的作用。
方法选取健康雌性食蟹猴6只笼养,3只为正常饲养的对照组,3只为除正常饲养外还给予口服7,8-DHF进行干预的实验组。注射显像剂99Tcm-TRODAT-1后分别于不同时间行SPECT/CT显像,取SPECT/CT图像中ST显示最清晰的连续横断层面勾画ROI,并选小脑(CB)作为参考本底,分别计算注射后1、3、4、5 h的ST/CB放射性摄取比值。统计学处理采用配对t检验。
结果实验组和对照组ST/CB比值均随时间延长不断升高,对照组注射后1、3、4、5 h的ST/CB比值分别为1.43±0.04、1.82±0.06、2.04±0.12、2.42±0.23,实验组分别为1.35±0.08、2.40±0.09、2.74±0.13、3.25±0.15。2组注射后1 h的ST/CB比值差异无统计学意义(t=2.57, P〉0.05),3、4、5 h时的ST/CB比值差异均有统计学意义(t值:2.77、2.87和2.92,均P〈0.05)。
结论99Tcm-TRODAT-1 SPECT/CT显像可用于评估7, 8-DHF对食蟹猴ST中DAT的激活作用。
Objective To assess the effects of 7,8-clihydroxyflavone (7,8-DHF) on the stfiatum (ST) in normal eynomolgus monkeys using 99Tcm-TRODAT-1 imaging. Methods A total of six healthy fe- male cynomolgus monkeys were included in this study. Three of them were fed with normal food ( control group) , and the other three were given oral administration of 7,8-DHF in addition to normal food ( experi- mental group). The SPECT/CT imaging was performecl at different time after 99Tem-TRODAT-1 injection. The [{OI of ST was drawn on images of 3 consecutive transverse slices that could be visualized best. The cer- ebellum (CB) was taken as the baekgrouncl reference area. The radioactivity uptake ratios of ST/CB at 1, 3, 4 and 5 h were calculated respectively. Paired-t test was used to analyze the data. Results ST radioac- tive uptake ratios showed continuing increase on the delay images. ST/CB uptake ratios of the control group at 1, 3, 4 and 5 h were 1.43±0.04、1.82±0.06、2.04±0.12、2.42±0.23, respectively, and those of the ex- perimental group were 1.35±0.08、2.40±0.09、2.74±0.13and3.25±0.15 respectively. There was no signifi- cant difference between the two groups at 1 h (t = 2.57, P〉0.05) , while ST/CB uptake ratios of the experi- mental group at 3, 4 and 5 h were significantly higher (t values: 2.77, 2.87 and 2.92, all P〈0.05). Conclusion 99Tcm-TRODAT-1 SPECT/CT imaging can be used to assess the DAT activation effect by 7, 8- DHF on ST of cynomolgus monkeys.
