摘要
目的探讨及分析透明细胞肾细胞癌(ccRCC)长链非编码RNA(lncRNA)的表达谱,并进行初步验证。方法利用高通量lncRNA芯片技术分别检测3例ccRCC组织及对应癌旁组织中lncRNA表达谱的差异,对芯片结果进行基因本体论(Gene Ontology)分析,Pathway分析,建立lncRNA与靶基因的基因网络图,并利用实时荧光定量RT-PCR技术对高通量lncRNA芯片检测结果进行验证。结果 ccRCC患者癌组织与癌旁对照样本差异表达的lncRNA共3 862条,其中1 619条上调表达,2 243条下调表达(差异表达倍数≥2.0,P<0.05)。荧光定量RT-PCR结果显示,筛选出的6个lncRNA,在87例ccRCC样本中的表达差异与芯片检测结果一致。结论ccRCC存在明显的lncRNAs差异性表达,这些lncRNAs及某些信号通路可能在肾癌发生和发展中发挥重要作用。
Objective To analyze and prelimiarily validate lncRNA expression profiles in clear cell renal cell carcinoma(ccRCC).Methods LncRNAs and mRNAs expression profiles in ccRCC samples compared with non-tumor(NT)samples were performed by high-throughput microarray.The interesting candidate lncRNAs were verified by real-time quantitative PCR.In addition,Gene Ontology(GO)and pathway analyses for differentially expressed mRNAs were performed using the standard enrichment computational method and a co-expression network of these coding-noncoding genes was constructed in our work.Results Among the 78 243 lncRNA transcripts examined,3 862 lncRNAs were found to be significantly differentially expressed(P 0.05,2.0-fold change)in ccRCC samples relative to their matched counterparts,with1 619 being up-regulated and 2 243down-regulated.The expression levels of 6lncRNAs examined by qRTPCR were shown to be highly consistent with the microarray data.Conclusion The results revealed lncRNAs expression patterns in ccRCC by microarray.These lncRNAs and its pathways involved may play a key role in the development and occurrence of ccRCC.
