摘要
目的比较不同生长年限川牛膝药材中葛根素和杯苋甾酮的含量差异,为其采收年限确定和质量控制提供依据。方法采用高效液相色谱法,色谱柱为Kromasil C18(250 mm×4.6 mm,5μm),检测波长243nm,流动相0.1%磷酸溶液-乙腈,流速1.0 m L/min。结果不同生长年限川牛膝药材中杯苋甾酮的含量高低依次为:3年(0.84±0.14 mg/g)>2年(0.65±0.09 mg/g)>1年(0.46±0.08 mg/g);葛根素的含量高低依次为:3年(21.41±10.21μg/g)>2年(10.08±7.96μg/g)>1年(6.97±4.12μg/g)。结论不同生长年限的川牛膝药材中两种有效成分含量有明显的差异(P<0.05)。因此,应以3年生的川牛膝采收入药最佳。
Objective To compare the differences of puerarin and cyasterone in Cyathulae radix with different growth years, and provide a reference for the harvest age and quality control. Methods The samples were separated on a Kromasil C18 column (250 mm×4.6 mm, 5 μm). Acetonitrile-0.1 % phosphoric acid solution was used as mobile phase, with a flow rate of 1.0 mL/min. The detection wavelength was set at 243 nm. Results The contents of cyasterone in Cyathulae radix with different growth years were in the order: 3 years (0.84±0.14 mg/g) 〉 2 years (0.65±0.09 mg/g) 〉 1 year (0.46±0.08 mg/g), and the contents of puerarin were in the order: 3 years (21.41±10.21μg/g) 〉 2 years (10.08±7.96 μg/g) 〉 1 year (6.97±4.12 μg/g). Conclusion The contents differences of the two effective components in Cyathulae radix with different growth years are significant (P〈0.05). The results indicate that optimal harvest age of Cyathulae radix is 3 years.
