摘要 目的采用细菌回复突变试验、体外染色体畸变试验和小鼠骨髓微核试验研究牡蛎水提液的遗传毒性。方法细菌回复突变试验中,牡蛎水提液设5000,2500,1250,625,312.5μg/皿5个剂量,计数在有无代谢活化系统时各剂量组回变菌落数;体外染色体畸变试验中,设5,2.5,1.25 mg/m L 3个剂量,染毒6,24 h,观察各组染色体畸变率;小鼠骨髓微核试验中,设2000,1000,500 mg/kg 3个剂量,每日给药1次,连续给药2天,观察各组微核发生率。结果牡蛎水提液各剂量组回复菌落数、染色体畸变率、微核率与阴性对照组比较,差异均无统计学意义(P>0.05),结果均为阴性。结论在本试验条件下,牡蛎水提液未见潜在的遗传毒性。 Objective To evaluate the genotoxicity of oyster water extract by bacterial reverse mutation, in vitro chromosome aberration and mice bone marrow micronucleus tests. Methods In bacterial reverse mutation test, the revertant colonies of five different dosage (5000, 2500, 1250, 625, 312.5 μg/plate) groups with or without S9 were scored. In the in vitro chromosome aberration test, the chromosome aberration rates of three dose levels (5, 2.5, 1.25 mg/mL) of oyster water extract treated groups after 6 h or 24 h of treatment were observed. In mice bone marrow micronucleus test, the micronucleus rates of three dose levels (2000, 1000, 500 mg/kg) of oyster water extract treated groups after two days of treatment (once daily) were observed. Results The differences between the number of revertant colonies, the chromosome aberration rates and the micronucleus rates of the oyster water extract treated groups and those of negative control group were not significant (P〉0.05), and the results were all negative. Conclusion No genotoxicity of oyster water extract was observed in this experiment.
