摘要
通过生理鉴定和分子鉴定的方法,对经2.0 mol·m L^(-1)锌溶液处理后的草珊瑚叶片的内生菌进行分离纯化,培养得到了草珊瑚内生细菌单菌落,对菌落的培养特征、革兰氏染色等生理生化结果,以及提取内生细菌的DNA,并通过PCR得到其16SrDNA序列,通过BLAST比对分析,利用MEGA软件建立发育树等分子鉴定手段,初步推断所分离得到的草珊瑚内生细菌为芽孢杆菌。
Through the method of identifying the physiological and molecular identification, the endophytic bacteriawere isolated and purified on the leaf of Sarcandra glabra with the 2.0 mol. mLl solution of zinc-tolerant liquid. The singlecolony of endophytic bacteria from Sarcandra glabra was isolated, purified and cultivated. With the method of identifying thephysiological including the colony culture characteristics, Gram staining, and extraction of DNA, 16S rDNA sequence byPCR, blast analysis, establishing phylogenetic tree by MEGA software, it is deduced that the bacteria from Sarcandra glabrais Bacillus.
